Abstract


Objective To observe mycophenolate mofetil (MMF) protective effect on the kidneys of diabetic rats , and to explore its mechanism of action . SD rats with unilateral nephrectomy two weeks were randomly divided into a control group (C group ) and the experimental group . The experimental group by intraperitoneal injection of streptozotocin (STZ) diabetes model , the control group injected with the same amount of buffer . After successful modeling experimental group were randomly divided into diabetic group (D group ) and diabetes treatment group (M group , 20 mg · kg -1 · d -1 ) . Observed after 12 weeks , their blood glucose (BG) , blood urea nitrogen (BUN) , serum creatinine (Scr) , renal hypertrophy index ( kidney weight the KW / weight BW) , creatinine clearance (Ccr) and 24-hour urinary protein ( 24Upro). Kidney tissue do conventional light microscopy and electron microscopy to observe the morphology . By immunohistochemical methods to detect kidney tissue of proliferating cell nuclear antigen (PCNA) , macrophage marker antigen (CD68) , intercellular adhesion molecule -1 (ICAM-1) and monocyte chemoattractant protein -1 ( MCP -1 ) protein expression. Renal tissue ICAM-1 and MCP-1 mRNA expression was detected by semi- quantitative RT-PCR method . Results Compared with the control group , the diabetic rats BG , KW / BW , 24Upro , BUN , Scr and Ccr significant rise (P lt; 0.05 or 0.01 ) ; the pathological results show that the the mesangial density and glomerular basement film thickness were significantly significant increase (P lt; 0.01); immunohistochemistry chemical testing results of renal tissue PCNA , CD68 , expression of ICAM - 1 and MCP-1 in protein expression were significant with raised ( P lt ; 0.05 or 0.01 ) ; semi- quantitative RT -PCR showed that renal tissue ICAM-1 and MCP-1 mRNA expression was significantly increased (P lt; 0.05 or 0.01 ) . Group in the treatment of diabetes , the indicators in addition to blood glucose were significantly inhibited (P lt; 0.05 or 0.01) . Conclusion In diabetic rat model , MMF reduce urinary protein to reduce glomerular hypertrophy , prevents mesangial cells , mesangial matrix and basement membrane thickening , which has a protective effect on diabetic nephropathy (DN) , which may be related lowered PCNA and CD68 expression , reduce monocyte / macrophages in the renal tissue of aggregation , inhibition of renal tissue ICAM-1 and MCP-1 expression .
