Abstract


Purpose of MCF-7 breast cancer cells as cell model to investigate Triptolide Cinobufotalin the cell proliferation , as well as the cellular oncogene c-myc transcription and translation effects . Method 1 , MCF-7 cells synchronized , with different concentrations of TP and CBC were cultured at different times . 2 , by MTT assay MCF-7 activity. 3 , the use of the RT-PCR method semiquantitative cell oncogene c-myc mRNA levels . 4 , the use of immunohistochemical methods semi-quantitative cell oncogene c-myc protein expression levels . Results 1, TP concentration 75-150μg/ml in the range , the time range of 8 ~ 24h could inhibit the proliferation of MCF-7 cells (P lt; 0.01), showed a good dose-response relationship and effect relationship ; significantly inhibited MCF -7 cellular oncogene c-myc mRNA levels and protein expression (P lt; 0.01), showed a good dose-effect relationship . 2, CBC at a concentration of 0.2g/ml (P lt; 0.05), 0.4g/ml (P lt; 0.01), within the time 8 ~ 24h significantly inhibited the proliferation of MCF-7 cells , showed a good dose-effect relationship and effect relationship ; significantly inhibited MCF-7 cells the oncogene c-myc mRNA and protein expression showed a good dose-effect relationship . 3 , the two combined no obvious synergies. Conclusion 1, TP can inhibit MCF-7 breast cancer cells proliferation and oncogene c-myc transcription and translation . 2, CBC can be inhibited MCF-7 breast cancer cell proliferation and oncogene c-myc transcription and translation . 3 , two kinds of traditional Chinese medicine combined fight breast cancer is no obvious synergies.
