Correlation between the Expression of Podocin mRNA and Oxidative Stress in Adriamycin-induced Nephrotic Rats

Abstract


Objective To investigate the rats with adriamycin proteinuria developing glomerular slit diaphragm molecule podocin mRNA expression and oxidative stress relationship. Methods 24 SD rats tail vein single injection of doxorubicin 7.5mg/kg established adriamycin, normal control group, 10 rats were injected with normal saline. 14, 21 days were sacrificed eight rats with adriamycin urine specimens and kidney samples, detection of 24h urine protein; renal cortical homogenates determination of malondialdehyde (MDA), superoxide dismutase (SOD) correlation analysis and total antioxidant capacity (T-AOC); situ hybridization staining and semi-quantitative RT-PCR assay podocin mRNA distribution and expression, and expression of oxidative stress indicators and podocin transcriptional level. (1) ADR injection 7d 24h urinary protein excretion compared with normal control group increased, but no statistically significant difference (p = 0.08) after ADR injection 14d urinary protein excretion was significantly higher than the normal control group (p lt; 0.01), 21d after injection further increased (P lt; 0.01). (2) ADR injection after of 7d renal cortex homogenate MDA content was significantly increased, the mean is 1.24 times the normal control group (P = 0.02), the ADR injection 14d is 1.30 times the normal control group (P lt; 0.01) , the 21d group of MDA content was significantly higher, 1.42 times the normal control group (P lt; 0.01); renal cortex of SOD in the ADR injection after 7 days of decline, but no significant difference compared with the control group (P = 0.15 ), 14d group of 89% of the normal control group (P = 0.029), the future continues to decline, the 21d group is of 83% of the normal control group (P = 0.001); renal cortex homogenate T-AOC the ADR injection 7d group and 14d were 96% and 80% of the normal control group, but no significant difference compared with the control group (P GT; 0.05), the future continues to decline, the the 21d group is of 77% of the normal control group (P = 0.036) . (3) in situ hybridization staining normal control group podocin mRNA is expressed mainly in the glomerular cytoplasmic; adriamycin nephropathy group with the progress of time, the number of positive cells increased significantly positive strength significantly increased; negative control glomerular and the renal tubules showed no blue particles. (4) Semi-quantitative RT-PCR results showed that ADR injection after the the 7d group of podocin mRNA expression compared with the control group did not change significantly (P = 0.086), 14d group and 21d group was significantly higher, are normal control group, 1.56 times and 1.87 times (P LT; 0.01) (5) The correlation analysis showed that podocin mRNA expression of renal cortical malondialdehyde content was positively correlated (r MDA = 0.443, p lt; 0.05), and superoxide dismutase and total antioxidant capacity was significantly negatively correlated (RSOD = -0.703, p lt; 0.01; rT-AOC = -0.467, p lt; 0.01). Conclusion nephrotic state podocyte molecules podocin mRNA abnormal expression of oxidative stress are closely related.
