Analysis of Differencially Expressed Proteins in Root from Puccinellia Teniflora under Nacl Stress

Abstract


Puccinellia tenuiflora, which is a herbaceous plant species, belong to the genus Puccinellia in Gramineae, it is also a good model to study the mechanism of salt-tolerant. We analyed the part of physiological indexes and differentially expressed proteins in root of Puccinellia tenuiflora. The main results include, (1) the proline content, the malondialdehyde content and the soluble sugar content were all inceased in the root; (2) the peroxidase(POD) activity, the catalase(CAT) activity, the glutathione reductase(GR) activity and the ascorbate peroxidase(APX) activity were all increased but superoxide dismutase(SOD) activity was decreased in root; (3) we found 104 differentially expressed protein spots which response salt stress in root, 53 spots were up-regulated, while 51 were down-regulated, differentially expressed protein spots main congregate in the range of pH5-5.5 and 30-70kD. Our results show that, membrane system in root of Puccinellia tenuiflora has been damaged after treated by 600mM NaCl for 24 hours, The root regulated the osmotic balance by accumulation of proline and soluble sugar. Meanwhile, the root started antioxidant defense system in order to scavenge ROS, so that it can cope with high concentrations of salt stress. In addition, changing in abundance of so many proteins with different isoelectric point and molecular weight showed that, roots of Puccinellia tenuiflora may enhance salt tolerant ability by regulating numerous metabolism pathways.
