Abstract

Günther (Cynoglossus semilaevis) is an economically important fish in China, it has a delicious meat, rich in nutrition, the advantages of rapid growth. Many studies have shown that polyploid fish, especially triploid fish than diploid better growth and reproductive traits, it is becoming fish breeding, fish trait improvement goals. As the research object Günther, cold shock induced triploid Günther, screening the best inducing conditions. In addition, Universal amplification our laboratory the circular spot Star flounder microsatellite primers in closely related species, the genetic polymorphism and plaice and the Konoha flounder wild populations. The main results are as follows: 1. Semi slippery tongue sole normal sperm and egg fertilization, after fertilization, at the appropriate time using cold shock inhibit its second polar body exhaust induction of Günther triploid, and explore the triploid induction of good conditions: 23 ° C water temperature after fertilization 4-5min, the fertilized egg on the 4 ° C seawater cold shock treatment, continuous time of 20min triploid induction rate was 35%. Cytometric DNA analysis relative content and chromosome preparation method, prove its triploid. Triploid chromosome analysis, identify the type of the male and female chromosomes the ZWW / ZZZ, the frequency of occurrence of 1:1, found no ZZW-type females, suggesting Günther Z cross between the chromosome and W chromosome and swap severely inhibited . 2 In this study, using the laboratory through enriched library - colonies in situ hybridization screening 216 pairs the circular spot Star flounder microsatellite primer spots in three closely related species strip star flounder, plaice and Konoha plaice The versatility amplification. With the primer of the amplified specific bands in the three species 137,95,90, the polymorphic primers number 82,35 and 26, respectively. Study, microsatellite marker primers the round plaques Star flounder case in Star flounder, (streaks Star flounder) higher versatility, the rest are in the flounder suborder (plaice and Konoha flounder) fish versatility relatively lower, and then verify that the the microsatellite sequence conservation with species genetic distance relationship. Plaice 20 polymorphic primers and Konoha flounder 18 polymorphic primers, respectively, its wild populations of the Yellow Sea region analysis of genetic diversity found in each of the sites in the plaice The number of alleles 5-14, the average number of alleles 9.5, observed heterozygosity and expected heterozygosity ranged from 0.1724-1.0000 and 0.5850-0.9226, there are six sites deviate significantly from Hardy-Weinberg equilibrium (p lt; 0.0025); number of alleles found in each site Konoha flounder 6-14, the average number of alleles 10.22, observed heterozygosity and expected heterozygosity range 0.3846-1.0000 and 0.6810, respectively. - 0.9286 7 sites deviate significantly from Hardy-Weinberg equilibrium (p lt; 0.002777). The results show that these microsatellite markers with high polymorphism information content, suitable for a variety of genetic analysis.
