Improvement of crop salt tolerance to utilize salinity-alkalinity soil is one of the important tasks in development of agriculture. Compared to NaCl salt stress, carbonate stress from inland salinity-alkalinity soil is more severe to plants and it’s mechanism of plant tolerance remains obscuree.In present study, we studied the maize seedling roots of three-leaf stage under 25mM Na2CO3 stress, with water treatment as control, to investigate the deferential proteomics profile by using 2-DE and MS. We analyzed the changes of 2-DE gel patterns of the root protein under Na2CO3 stress with deferent treatment time such as 5h, 24h and 72h and then we identified by MS the protein spots in the gels that were changed significantly in comparison with the control. The results of 2-DE analysis indicated that 1291 protein spots were found to exhibit different dynamic patterns on 2-DE gels in response to 25mM Na2CO3 stress and 12 of them were identified successfully using MALDI-TOF-TOF MS. These salt responsive proteins, besides one protein of unknown function, were involved in regulation of metabolism, transcription, cell cycle, signal transduction, cell defense, defense/interaction with environment by MATDB database survey and the related references, which show the response to salt stress involves many signal and metabolism pathway in plants. We discussed the possible metabolism of the gene regulatory network in response to salinity-alkalinity stress according to our results. The information obtained provided important inshights into understanding the mechanisms of salinity-alkalinity tolerance in plants.
