Abstract


ObjectiveTo identify some differentially expressed proteins in renal cell carcinoma patients, proteomics technologies were applied into research compared with RCC tissue and normal-adjacent kidney, serum from RCC patiens and benign kidney mass.MethodsPartⅠTotal proteins of RCC tissue and paired normal tumor-adjacent kidney tissue were separated by two-dimensional gel electrophoresis, differential proteins were identified by MALDI-TOF/TOF-MS;PartⅡMALDI-TOF MS combined with magnetic beads were applied to the study of a cohort of serum samples from renal cell carcinoma patients and benign kidney mass, Proteomic fingerprinting of serum were identified and analyzed by Biomarker Wizard Software and Biomarker Patterns Software.Result:PartⅠA stable protein expression profile of RCC tissue is successfully set up compaired with normal-adjacent kidney tissue. RCC tissue has 10 differently express proteins and successfully identifies 8 protein using MALDI-TOF/TOF-MS, chromosome 21 open reading frame 33 and ferritin light subunit with up-regulated respectively, AnnexinⅣ, aminoacylase-1, ATP synthase, L-Lactate Dehydrogenase H Chain and histone H2bl with down-regulated respectively.PartⅡSeven discriminative peaks were identified between group RCC and benign mass. A decision tree model with 4 discriminative peaks with the m/z 2945.35、m/z15340.8、m/z6984.51、m/z5819.23 was generated by Biomarker Patterns Software, and a sensitivity of 83.0% and specificity of 84.6% were observed. A new group of patients with RCC and benign kidney mass were introduced to verify the model with sensitivity of 80% and specifity of 81.8%. ConclusionProteomics technology is a powerful tool for study of renal cell carcinoma;Cell metabolic enzymes, cytoskeletal proteins, and many other proteins may be involved in the pathological process of renal cell carcinoma;MALDI-TOF MS combined with magnetic beads for detecting serum protein peaks could be applied to renal cell carcinoma research, and establish the diagnosis model;Analysis of these differentially expressed proteins may contribute to tumormarker discoveries of renal cell carcinoma.
