Abstract


Kidney diseases such as IgA nephropathy (IgA nephropathy, IgAN), glomerulonephritis, is a common primary glomerular disease, some patients may progress to end-stage kidney disease, endangering people's health. Transforming growth factor β1 (transforming growth factor beta 1, TGF-β1), integrin-linked kinase (integrin-linked kinase, ILK), β1 integrin (β1 integrin) and the peroxisome proliferator bribes material activated receptor γ ( peroxisome proliferators-activated receptor gamma, PPARγ) has a very close relationship with the progression of renal disease. At the same time, the extracellular matrix (of extracellular matrix, ECM) metabolic imbalances, increased synthesis or degradation of the ECM protein components resulted in the reduction of the excessive accumulation of the main pathological features of glomerulosclerosis. Fish oil is extracted from the deep-sea fish rich in n-3 polyunsaturated fatty acids (n-3 polyunsaturated fatty acid, n-3 PUFA), the main active ingredient is eicosapentaenoic acid (eicosapentaenoic acid, EPA) and docosahexaenoic acid (docosahexaenoic acid, DHA). Fish oil in addition to the classic role of antithrombotic, vasodilatory, and lipids, anti-tumor, also has a renoprotective effect and slow the progression of kidney disease. Experimental injury model of rat mesangial cells induced by lipopolysaccharide (lipopolysaccharide, LPS) (glomerular mesangial cells, GMCs) observed EPA and DHA GMCs of MMPs, PPARγ, ILK, β1 integrin, TGF-β1 expression affect explore the possible protective mechanism of injury GMCs, provide a theoretical basis for clinical application. Methods GMCs, conventional logarithmic growth phase cultured for 24 h, serum-free cultured for 24 h, the cells synchronized in the G0 phase. Divided into: normal control group; LPS (model group, LPS10.0mg / L) group; EPA (10μmol / L, 100μmol / L) group; DHA (10μmol / L, 100μmol / L) group, in which the drug group containing LPS medium. Each group were cultured for 24 h and 48 h after the index determination. 1 by ELISA in observed GMCs TGF-β1 secretion EPA and DHA on LPS stimulation; 2 GMCs MMP-2, 9 activity of EPA and DHA on LPS stimulation by SDS-PAGE gelatin zymopraphy analysis; 3 observed by Western Blot EPA and DHA on LPS-stimulated GMCs PPARy and ILK protein expression; 4 RT-PCR was used to observe the EPA and DHA on LPS-stimulated GMCs PPARy, ILK, β1 integrin and TGF-β1 mRNA expression in; results of EPA, DHA LPS-stimulated GMCs TGF-β1 LPS stimulation, GMCs synthesis and secretion of TGF-β1 mRNA expression increased EPA and DHA than the normal control group decreased LPS-induced the GMCs TGF-β1 synthesis and secretion and mRNA expression can be. 2 EPA, DHA increased in LPS-stimulated of GMCs were expression of MMP-2, 9 activity in LPS-stimulated GMCs of MMP-2, 9 synthesis and secretion compared with the normal control group decreased GMCs; EPA and DHA LPS-induced expression of MMP-2, 9 synthesis and secretion 3 EPA, DHA the GMCs PPARy mRNA and protein expression can affect LPS LPS stimulation GMCs PPARy expression decreased; EPA and DHA increase LPS-induced GMCs PPARy mRNA and protein expression. 4 EPA, DHA LPS stimulation the GMCs ILK expression LPS increase GMCs ILK mRNA and protein expression; EPA and DHA allows decreased LPS-induced GMCs ILK mRNA and protein expression. The the 5 EPA, DHA stimulate GMCsβ1 integration on LPS mRNA in LPS increases GMCsβ1 integrin mRNA expression; EPA and DHA can LPS-induced GMCsβ1 integrin mRNA expression decreased. Conclusion EPA and DHA reduce LPS-induced GMCs TGF-β1 protein and gene expression. 2 EPA and DHA GMCs to promote LPS-induced secretion of MMP-2, 9. 3 EPA and DHA can promote LPS-induced GMCs PPARy protein and gene expression. 4 EPA and DHA reduce LPS-induced GMCs (31 integrin and ILK protein and gene expression.
