Acupuncture preconditioning on rat model of Alzheimer Wnt signal transduction pathway in rats

Abstract


Objective To establish a rat model of AD to explore acupuncture pretreatment on senile dementia model rats Wnt 1 signal transduction pathway reveals mechanism of acupuncture prevention of AD. Methods in the rat hippocampus bilaterally injected Aβ 25-35 , create an AD rats were randomly divided into normal group, sham operation group, model group, pre-moxibustion group, the pre-EA group , pre-electroacupuncture plus moxibustion group, a total of six groups, each group 20. By pre-EA and pre-moxibustion rats Baihui and Shenshu 28 days, rats were observed wnt signal transduction related indices. Morris water maze test behavior, immunohistochemistry and Western blot hippocampus axis and β-catenin protein by enzyme-linked immunosorbent assay of protein phosphatase 2A and glycogen synthase kinase-3β activity through real-time Wnt1 quantitative PCR to detect gene expression, and through micro electron microscopy ultrastructural changes in the hippocampus. Results 1 found water maze behavioral model of learning and memory abilities of rats obvious damage, the average incubation period, the proportion of through the central region, the average swim path distance with the normal group and sham operation group were significantly different (p lt; 0.01), while the pre-electro-acupuncture and moxibustion expected to exert significant preventive and improvement, which pre efficacy of acupuncture and moxibustion best. The pre-moxibustion group, the pre-EA group, electro-acupuncture and moxibustion pre hippocampal Wnt1 signals have different levels of enhancement. Conclusions 1. Senile dementia of the basic pathogenesis marrow deficiencies, God Machine apraxia, acupuncture rule-Yijing fill the marrow, Stasis resuscitation. Baihui and shenshu can be used as the main point of choice for prevention and treatment of disease. (2) Pre-electro-acupuncture and moxibustion for pre-AD neurological function of rats with a significant protective effect, effectively improve the AD model rats learning and memory function, reducing neuronal damage, in order to pre-electro-acupuncture and moxibustion combined use of pre- best effect. 3 Pre-electro-acupuncture and moxibustion can significantly decrease the pre-AD model Axin expression in hippocampus, increased β-catenin expression, reducing the activity of GSK-3β, increased PP2A activity and enhance Wnt1 gene expression. 4 Acupuncture prevention of Alzheimer's mechanism of action may regulate Wnt signaling pathway, affecting learning and memory-related enzymes and protein expression in the hippocampus play a role.
