Abstract

Objective: To establish an animal model of rat spinal cord injury. Observed weight support treadmill training combined with electro-acupuncture in the treatment of spinal cord transection injury the rat injury site of nerve growth associated protein (GAP-43), neuronal apoptosis-related protein (Caspase-3). Explore weight loss recovery mechanism walk training combined with electro-acupuncture in the treatment of spinal cord injury in rats function. Methods: 85 Clean-level six-week-old healthy adult Wistar rats weighing 180-220 g, all female. Were randomly divided into five groups, the blank group 5, sham group, the control group, electro-acupuncture group weight support treadmill training the combined EA group (needle Kang group), 20 in each group. Spinal cord injury model was successfully manufactured the first three days of the EA group and needle Kang group treated accordingly. A total of 28 days of treatment. 3 days, 7 days, 14 days, 21 days, 28 days on rats in each group BBB score, and rats in each group drawn from three rats in the other four groups of rats in the control group, select , do paraffin sections of whichever injury District spinal cord tissue by immunohistochemistry and image analysis were observed GAP-43, caspase-3 expression changes. Results: ① spinal cord transection injury rats walking dysfunction obvious, spinal cord injury model modeling success. GAP-43 was no expression in normal adult rat spinal cord, spinal cord injury, spinal cord increased expression of GAP-43 spinal cord injury on day 3 expression was significantly increased 7 days expression reached a peak and then decreases. GAP-43 content of the same point in time comparison, drawn from outside of the 3 days, the needle the Kang group of more than the EA group were significantly different (p lt; 0.05). EA group than the control group, also a significant difference (p lt; 0.05). ② Caspase-3 in normal adult rat spinal cord shows little expression in the spinal cord after spinal cord injury increase in the expression of Caspase-3 expression was significantly increased spinal cord injury 3 days, 7 days expression reached a peak and then decreases. Compare the same point in time Caspase-3 content, except 3 days drawn, needle Kang group less than the EA group were significantly different (p lt; 0.05). EA group than in the control group also had a significant difference (p lt; 0.05). ③ successful modeling three days needle Concord group, EA group and the control group each rats BBB score showed an increasing trend, but at the same time point BBB scores between needle Kang group was better than the EA group significant differences (p lt; 0.05). EA group than the control group also had a significant difference (p lt; 0.05). Weight support treadmill training combined with electro-acupuncture treatment with simple electro-acupuncture treatment can promote an increase in GAP-43 content in the spinal cord of the rat damage zone, weight loss walk training combined with electro-acupuncture treatment most significant conclusions: 1. Weight support treadmill training combined with electro-acupuncture treatment with simple electro-acupuncture treatment can inhibit Caspase-3 levels within the spinal cord of the rat damage zone increased weight support treadmill training combined with electro-acupuncture treatment is most significant. 3 weight support treadmill training combined with electro-acupuncture treatment alone EA treatment of spinal cord injury in rats showed a stable upward trend in its limb BBB rated weight support treadmill training combined with electro-acupuncture treatment of spinal cord injury in rats, their limbs BBB score liter high than the simple electro-acupuncture treatment is more obvious.
