Abstract


In recent years, with the rapid development of tissue engineering , tissue engineering skin has begun to extensively in clinical use. The current source of seed cells for tissue-engineered skin is skin dermal fibroblasts and epidermal keratinocytes , skin appendages such as hair follicles, sebaceous tissue-engineered skin constructed therefrom , there is still a large organizational structure and normal skin tissue differences. Hair follicle cells play an important role in the repair of skin wounds , can be used as a source of seed cells involved in skin wound repair after using hair follicle cells at home and abroad to build tissue-engineered skin fewer reported in the literature . In this study, in the isolation and culture of outer root sheath cells and dermal sheath cells and dermal papilla cells based on the use of tissue engineering techniques , other than root sheath cells and dermal sheath cells as seed cells , preparation of a new tissue-engineered skin , and successful tissue engineering full-thickness skin defects of the skin of the animal model of transplantation experiments testing the biological activity of the tissue-engineered skin . At the same time , further attempts to build the hair follicles of the tissue-engineered skin adnexal tissue-engineered skin containing a more in-depth study . Specific experiments are as follows : 1 , a simple and efficient training methods to get a lot of growth in good condition the outer root sheath cells , dermal sheath cells and dermal papilla cells : cell-derived newborn SD rats , using mechanical method extractor complete growth early hair follicle digestion method for dermal sheath cells and outer root sheath cells , and tissue explant culture of dermal papilla cells . 2 , tissue-engineered skin culture system designed in the laboratory , the first dermal sheath cells as seed cells to construct tissue engineered dermal complete hair follicles in the dermis of the tissue engineering composite formation epidermis
