Virus Inactivation in Red Blood Cells by Photochemical Method and Related Mechanisms

Abstract


In order to find a new phenothiazine photosensitizer that can be used in virus inactivation in red blood cells (RBCs) , a series of phenothiazine photosensitizers were tested, the relationship between their structure and function was discussed。 Among these new photosensitizers, M007 was the best one. Compared with MB and DMMB, which have been used in virus inactivation in RBCs, M007 cause less damage to RBCs than MB, but similar with DMMB. Results of virus-inactivation by M007 were more reproducible than DMMB in different units.Using M007 as the photosensitizer, the four parameters of phototreatment (revolution of sample stage, time for dark incubation, Hct and sample thickness) were investigated and optimized. Under this optimized condition, M007 phototreatment inactivated 6.2 ± 0.3 Log10 bacteriophage Phi6, >6.5±0.6 Log10 VSV, >5.3±0.5 Log10 HSV-1, >9.9±0.4 Log10 bacteriophage R-17.
