Transforming growth factor-β (TGF-β) comprises a large family of pluripotent growth factors which hare been implicated in the diverse living processes including the growth, prolifereation, differentiation, and programed cell death (or apoptosis) in normal, transformed, and cancer cells. Ovarian cancer remains the most lethal disease among the gynecological malignancise at present. The relationship between TGF- 3 and ovarian cancer was investigated in this paper.From the malignant ascites of patients with ovarian cancer, we established two epithelial ovarian cancer cell lines, COC1 and COC2, as well as COC1- and COC2-bearing nude mice models, COC1n and COC2n.It was observed that the serum-free conditioned medium SFCM) from COCl and COC2 could separetely stimulate COCl or COC2 proliferation, in vitro culture, in an autocrine manner, and COC2 cells, incubated in different sites of the smae nude mice, could promote the growth themselves in defferent incubated sites each other. COC1- and COC2-patients ascites , from whom COC1 and COC2 were established, COC1n and COC2n sera, SFCMl and SFCM2 were able to inhibit phytohemagglutinin-and interlukine-2-inducing peripheral blood mononuclear cell (PBMC)proliferation.The abovmentioned SFCM effects might be partially blocked by anti-TGF-β antibody.It was also shown that both SFCM1 and SFCM2 could promote TGF-β indicator, NRK-49F cell, colony formation in semisolid culture, and TGF- β mRNA was detected by slot blot and cellular hybridization in situ.
