Studies on Polymorphisms on Exon and Intron of BMP7 Gene and Their Associations with Reproductive Traits in Pigs

Bone morphogenetic protein 7 is a member of the family of bone morphogenetic proteins. This multifuctional protein also plays an important role in formation of bone and cartilage, and is also involved in the regulation of mammalian reproduction. No reports on the genetic diversity analysis have been found so far in the studies of porcine BMP7 gene, thus the present study took gene as a target gene for the pig reproductive traits. First, a DNA pool was build using DNA samples from more than 100 sows of different breeds, and sequenced to screen the putative SNPs occurred in the exon and intron region of porcine BMP7 gene. Then after, the PCR-RFLP was employed to genotype the total 258 sows and gilts including 25 Landrace, 148 Large white, 85 Duroc at the 2 microsatellite loci (STC, STA) and 4 SNP loci (T98C, T143C, T20C, T34C, T75C) found. The main results are as follows:(1) A total 92 polymorphic loci, including of 5 insert-deletes and 81 signal nucleotide polymorphisms and 6 microsatellites, were pick out for the first time in the studies of this gene.(2) Analysis results demonstrate that there has no polymorphism. 2 of the six microsatellite existing intron1 had been tested and analyzed by PAGE electrophoresis in polymorphism. Different genotypes in 2 microsatellite loci had been analyzed in the relevance to reproductive performance. Analysis results demonstrate that there are two genotypes on locus STC, which are low polymorphism (0.231). There also exist five genotypes on locus STA, which are moderate polymorphism (0.391). It had been found over Correlation analysis that STC locus has little significant effect on characters associated with breeding of pig, to the contrary, STA locus has significant effect on total number born (TNB) (p<0.05). Individuals with AC type perform better than ones with other genotypes in all characters associated with breeding. With two loci merger genotypes, individuals with AB/AC type have difference with other genotypes in all characters associated with breeding. The findings show that there exists certain interaction between this two microsatellite loci. It was found while grouping STC locus that there exist two noninterest strips except two interest strips on the opening side. Over Initial speculation, the two noninterest strips maybe heteroduplex forming in the process of PCR of Heterozygote.(3)The genotyping for the 2 variations found reveled that each of these loci occurred in the population studied with the full 3 genotypes of 5 sites, T98C was dominated by the alle A and genotype AB, with a alle frequency and genotype frequency of 0.657 and 0.500, respectively. The further examination showed that this locus coincided with the Hardy-Weinberg equilibrium, with a polymorphic information content(PIC) ,heterozygosity (He)and effective alle number(Ne) of 0.349,0.451 and 1.821 in the population of interest, respectively. At the locus T143C, the dominated alle and genotype were D (0.607) and DD (0.426), respectively. Whereas, this locus was not in the balance of Hardy-Weinberg, and PIC, He and Ne were 0.363, 0.477 and 1.913, respectively.At the locus T20C, the dominated alle and genotype were E (0.777) and EE (0.647), respectively. Whereas, this locus was not in the balance of Hardy-Weinberg, and PIC, He and Ne were 0.286, 0.346 and 1.530, respectively.For another locus T34C, the dominances were G and GH, with the frequencies of 0.624 and 0.450, respectively. This locus coincided with the Hardy-Weinberg equilibrium, and the PIC was 0.359, the He 0.469 and Ne 1.884.At the fifth locus T75C, the alle J was dominant to K, while the JJ was dominant to the remaining genotypes. The frequencies of J and JJ were 0.822 and 0.713, respectively. The locus A-1027G did not meet the balance of Hardy-Weinberg, and PIC, He and Ne were 0.250, 0.293 and 1.414, respectively.(4) The subsequent association analysis of these 5 loci with the corresponding reproductive traits demonstrated that T98C, T34C, T75C have little signification difference in total number born (TNB), number born alive (NBA), born litter weight, Litter weight at 21 days.The results of relevance analysis on T143C and T20C show that the T143C locus perform significant difference (p = 0.025 <0.05) in weight with 21d age, which site works mainly as add effect. In its genotypes individuals of CC type weight 3.484 kg more than individuals of DD type in Litter weight at 21 days, one data means significant difference (p < 0.05)between them. T20C locus has significant difference (p < 0.05) performance in pigs’ number born alive (NBA) and born litter weight, it performs very significant difference (p = 0.000 < 0.01) in Litter weight at 21 days. There exists super dominant effect between E, F alleles. This site works mainly as dominant effect, which plays a role in the production of individuals of EF genotype have 0.429 head number born alive (NBA) more than individuals of EE genotype (p < 0.05), meanwhile individuals of EF genotype have 0.754kg born litter weight more than individuals of EE genotype (p < 0.05), and individuals of EF genotype also have 4.385 kg in Litter weight at 21 days more than individuals of EE genotype (p< 0.01). So, heterocyst in T20C site could be consider to use while pig breeding according to our need in total number born (TNB ) , born litter weight and Litter weight at 21 days.Therefore, it can be reasonably concluded as follows:(1)The exon and intron region of porcine BMP7 is rich in the genetic variations, implying a great role in the regulation of this gene as well as the phenotypes related to the reproduction.(2)Though the further assertation is needed, the loci STA, T143C and T20C seem to be a possible molecular marker for the selection of corresponding traits in the swine breeding.
