Abstract
Objective: The superparamagnetic chitosan plasmid(pDsVEGF165Red1-N1)gelatin microspheres (SPCPGM) were prepared and filling into the porosity cages of biological activity artificial bone. The ability of the controlled release and local concentration of the plasmid DNA were investigated in vitro under a static magnetic field or an oscillating magnetic field. In vivo, the efficiency of slow release, controlled release, local concentration and delivery of the plasmid DNA for enhancement of angiogenesis and osteogenesis were evaluated under a static magnetic field and / or an oscillating magnetic field to know whether it could enhance local angiogenesis and osteogenesis and discuss value of clinical application in the future.Methods:⑴The superparamagnetic Fe3O4 chitosan nanoparticles (SPFCN) were prepared by a chemical co-precipitation, the appearance and size of the nanoparticales was surveyed with a Transmission Electron Microscope, The grain distributeon, zeta potentials and magnetic properties of the nanoparticles were determined with a Laser Particle Size Analyzer, a Malvern Zetasizer and a Magnetic Balance, respectively.⑵The recombinant eukaryotic expressive plasmids that can expresse the fusion protein of red fluorescent protein and
