Abstract


Cyclooxygenases(COXs) exist in at least two isoforms, COX-1 and COX-2. COX-1 expresses consistently in normal tissues. However, COX-2 could express only when stimulated by various factors, including cell factors, endotoxin, carcinogen, and so on. The level of COX-2 is so low that it is difficult to be detected in normal tissues. Researches have found that COX-2 highly expressed in a lot of tumor tissues, and participated in the occurrence and development of tumor through promoting cell proliferation, inhibiting cell apoptosis and accelerating tumor angiogenesis, and so on.RNA interference(RNAi) is a powerful gene tool targeting post-tra- -nscriptional gene silencing, which has recently been used to silence gene expression in various species. In our research, we designed and constructed vector-borne small interference RNAs targeting COX-2, consequently rese- -arched the inhibitory effect of COX-2, the condition of cell growth and the change of signal transduction. Our research might offer a new approach for gene therapy in gastric carcinoma.
