1 Copied the diet-induced obesity and insulin-resistance rat modelObjective: To copy the diet-induced obesity and insulin-resistance (IR) rat model to prepare for subsequent experiment.Methods: eighty-two male SD rats were randomly assigned into base-diet control group (Bas-C, n=10, fed with basal forage) and high-fat and high-sucrose diet group (HFS, n=72, fed with high-fat and high-sucrose forage). 8 weeks later, screening model rats: ① The rat in HFS which fasting weights was greater than the mean fasting weight plus 1.4 times of standard deviation of Bas-C was chosen as obesity model. ② The rat in HFS which homeostasis model assessment of IR (HOMA-IR) was greater than the mean HOMA-IR plus 1.4 times of standard deviation of Bas-C was chosen as IR model.Results: the achievement ratio of model copy in obesity and insulin-resistance rats was 58.3% and 66.7% respectively.2 The comparison of susceptivity to diet-induced obesity and insulin resistance in ratsObjective: To understand the difference and the mechanisms on susceptivity to HFS diet-induced obesity and IR in rats.Methods: The model copy methods, the model assessment methods on obesity and IR and the treatment of base-diet control group (Bas-C) equal to part 1. Rats in HFS (except SIF treatmented rats) were assigned into 4 groups: nonobesity and nonIR group (nOb+nIR, neither accordance with obesity nor IR standard), obesity + IR group (Ob+IR, accordance with obesity and IR standard), obesity+ nonIR group (Ob+nIR, accordance with obesity but not with IR standard), nonobesity+IR group (nOb+IR, not accordance with obesity but accordance with IR standard ). Bas-C rats were fed with basal forage and the other rats were fed with HFS forage continually. After 4 weeks, samples were harvested after overnight fasting.
