Abstract


“Relation among the herbs, formula, effects and syndrome”is an important question in determination of treatment based in pathogenesis obtained through differentiation of symptoms and signs and Formulaology in TCM..The critical question in formulaology is effective principle,which involves in the herb dosage, compatibility,dosage form,sume methods and syndrome.When we make up formula,we must consider the effective object,dosage and compatibility,in this way the herbs can thoroughly educe the clinical effects.In this topic,the“Relation among the herbs, formula, effects and syndrome”is the question background,the effect of Bupleurum root on SHUGANJIEYU is the incision point.To research the effects of different dosages Bupleurum root on GANYU model rars,we will search the dose-effect relationship of Bupleurum root on SHUGANJIEYU.Based on this research ,we will compare with the Bupleurum root-baical skullcap root, Bupleurum root-chuanxiong and Bupleurum root-astragali radix on SHUGANJIEYU effects.We aslo explore the effects of Bupleurum root on rats with diffent funcuion states.This topic is very significant to probe the compatibility on dosage and herb selection,it is also important to argument the clinical experience of Bupleurum root and reveal the modern connotation of TCM..Research 1: the assaying of saikoside a in Bupleurum root and preparationMethods:Used the HPLC to determine the content of saikoside a in Bupleurum root material,the preparations of Bupleurum root, the Bupleurum root-baical skullcap root, Bupleurum root-chuanxiong and Bupleurum root-astragali radix,which were extracted by water.Results:The content of saikoside a in Bupleurum root material is 3.335mg/g, in the preparations of Bupleurum root, the Bupleurum root-baical skullcap root, Bupleurum root-chuanxiong and Bupleurum root-astragali radix are 0.441mg/ml, 1.024、0.807和0.693mg/ml.Conclusions:After compatibility,the dissolution of saikoside a was increased. Research 2:Study on the effects of dosage on SHUGANJIEYU of Bupleurum root Methods:Sieved by the experiment of open-field-test,the male rats were assigned into normal group, the model group, the CHAIHUSHUGANSAN group(Chsgs group 8.5g/kg), the Bupleurum root different dosage groups( 0.27、0.81、2.43、4.86、9.72g/kg),According to body surface area,they were 1/3、1、3、6、12 times by 9g/d,8 rats in every group.Except the normal group,the other rats were binded in binding tanks for 3 weeks,3 hours/d(8:00AM-11:00AM).The body weight were messured every week.From the second week the rats were administrated according to the dosages,the normal group and model group were administrated the distilled water(1ml/100gBW) for 14 days.On the 0,7,21th day,the ethology were determined for 5 minutes by the open-field-test.After the end administration 30 minutes,the rats were anesthetized by 10% hydral,the blood were taken by abdominal aorta,separated the plasma and serum,measured the content of VIP,SP,and ACTH,COR.Then the hippocampus were splited quickly on ice,measued the weight,preserved in nitrogen Canister,determined the monamine transmitter.Results:1.the effects on body weight in stagnation of the liver qi model rats:Compared with the normal group,the binded rat body weight increased significant slowly(P<0.05); Compared with the model group,the rat body weight increased quickly, which were administrated for 2 weeks,the Chsgs group and three times group increased obvious quickly(P<0.05).2.the effects on ethology in stagnation of the liver qi model rats: Compared with the normal group,the rats which were binded for one week,appeared increasing tendency in creep;Binded 3 weeks,the square numbers in creep and the frequence in straight were obviously decreased(P<0.05).Compared with the model group,the rats administrated the preparation were all increased the square numbers in creep and the frequence in straight, the Chsgs group ,1 and 3 times group rats significantly increased the square numbers in creep(P<0.05).3. the effects on ACTH and COR in serum in stagnation of the liver qi model rats: Compared with the normal group,the content of ACTH was obviously descended,the content of COR was significantly increased in model rats,which were binded for 3 weeks. Compared with the model group,successive administration 2 weeks,the administration group could enhance the content of ACTH, the 1,3,6 times group were obvious (P<0.05).The 1 times group could manifestly decreased the content of COR (P<0.05).4. the effects on VIP and SP in plasm in stagnation of the liver qi model rats: Compared with the normal group, the content of SP was obviously descended,the content of VIP was significantly increased in model rats,which were binded for 3 weeks(P<0.05). Compared with the model group, the administration groups could decrease the content of VIP in plasma,the 1,3,6,12 times group were obvious (P<0.01).5.the effects on monamine transmitter in hippocampus in stagnation of the liver qi model rats: Compared with the normal group,the content of NA and 5-HT in hippocampus in model rats were obviously increased,which were binded for 3 weeks. Compared with the model group, all of the Bupleurum root groups administrated for 2 weeks could obviously descended the NA,the 3,6,12 times groups could significantly decreased the content of 5-HT(P<0.05). Conclusions:Every dosage group could improve the biological parameter in stagnation of the liver qi model rats.The optimal dose of bupieurum root was 9-27g/d on soothing the liver and relieving depression..Research 3:Study on the effects of compatibility on soothing the liver and relieving depression of Bupleurum rootMethods:According with the results of research 2, 13.5g/d was the optimal dose of bupieurum root. the Bupleurum root-baical skullcap root, Bupleurum root-chuanxiong and Bupleurum root-astragali radix were prescripted according to 4:3. Sieved by the experiment of open-field-test,the male rats were assigned into normal group, the model group, Bupleurum root group(BR group, 1.22g/kg), Bupleurum root-chuanxiong group(BC group,2.13g/kg), the Bupleurum root-baical skullcap root group(BB group,2.13 g/kg) and the Bupleurum root-astragali radix group(BA group, 2.13 g/kg), 10 rats in every group. Compared with the model group, the administration groups could decrease the content of VIP in plasma,the BA group was obvious (P<0.01);BR, BC group could significantly increase the content of SP (P<0.05).5.the effects on monamine transmitter in hippocampus in stagnation of the liver qi model rats: Compared with the normal group,the content of NA and 5-HT in hippocampus in model rats were obviously increased,which were binded for 3 weeks. Compared with the model group, all the groups administrated for 2 weeks could obviously descended the NA and the 5-HT,the BC and BB groups were significantly (P<0.05).Conclusions: Every group could improve the biological parameter in stagnation of the liver qi model rats. The best prescribion was the Bupleurum root compatibilite with the chuanxiong. Research 4:Research on the effects of Bupleurum root on diffent functional states in ratsMethods:The male rats were divided into the normal group,model group, stagnation of the liver qi model +Bupleurum root group(BR group),normal+Bupleurum root group(NB group),10 rats in every group. Compared with the model group,successive administration 2 weeks, the BR group was increased the content of ACTH obviously (P<0.05) and decreased the content of COR manifestly (P<0.05).But the NB group had no effects on the contents of ACTH and COR.4. the effects on VIP and SP in plasm in diffent functional states rats: Compared with the normal group, the content of SP appeared descended tendency,the content of VIP was significantly increased in model rats,which were binded for 3 weeks(P<0.05). Compared with the model group, the BR group could decrease the content of VIP in plasma,and significantly increase the content of SP (P<0.05).But the NB group had no effects on the contents of VIP and SP in plasma..5.the effects on monamine transmitter in hippocampus in diffent functional states rats: Compared with the normal group,the content of NA and 5-HT in hippocampus in model rats were obviously increased,which were binded for 3 weeks. Compared with the model group, the BR group appeared the descended tendency in the NA and the 5-HT. Compared with the normal group,the NB group could obviously increase the the contents of NA and the 5-HT. in hippocampus in normal rats.Conclusions:In the stagnation of the liver qi state,the Bupleurum root could show the effects on soothing the liver and relieving depression ,the effects on the normal states must be the therapeutic action or the toxic action.From all the results, we could find that the optimal dose of bupieurum root was 9-27g/d on soothing the liver and relieving depression.The best prescribion was the Bupleurum root compatibilite with the chuanxiong.on soothing the liver and relieving depression. In the stagnation of the liver qi state,the Bupleurum root could show the effects on soothing the liver and relieving depression, the effects on the normal states must be the therapeutic action or the toxic action.
