Abstract


Molecular imprinting technique is an analog three-dimensional configuration spaceof antigen-antibody complement the development of new biometric technology, smallmolecule imprinted polymers have been very mature. In contrast, biologicalmacromolecules imprinting in meaning of the Purification is more important. However,for the complex structure of biological macromolecules, molecular weight, in and out ofthe process of imprinted cavities are subject to certain obstacles.In addition, the structureof biological macromolecules are often susceptible to environmental, it put forward newdemands of the polymer preparation conditions. For the MIP in terms of biologicalmacromolecules, molecular imprinted microspheres and nanoparticles thanks to molecularimprinting with a large surface area, fast mass transfer rate, etc, can greatly improve theadsorption capacity of MIP and adsorption efficiency.This study explored the method of preparation of Biological macromoleculesimprinted thermo-sensitive nanogel, and temperature-sensitive gel successfully graftedonto nano-silica surface, achieved in nano-silica surface imprinting of biologicalmacromolecules. The main contents include:1) Conditions of thermo-sensitive nanogel preparation conditions were explored, at50℃and30℃,we got a suitable temperature phase transitionthermo-sensitive nanogel, provides the conditions for the imprinting of BSA andHRP.2) The template BSA and HRP were successfully imprinted by thermo-sensitivenanogel. For BSA-MIP, the adsorption of BSA (9.6mg/g) was significantlyhigher than NIP (1.8mg/g); For HRP-MIP, the adsorption of HRP (1.4mg/g)was significantly higher than NIP (0.8mg/g), the adsorption of HRP at30℃forMIP was significantly higher than at4℃, this also reflects the thermo-sensitive molecularly imprinted polymer selective adsorption and well control of thetemplate.3) Nano-silica were introducted into MIP preparation in as the of separation media.After nano-silica surface modification, template protein HRP(0.1μg/g) weresuccessfully fixed onto the surface, and successfully grafted to achieve athermo-sensitive gel for imprinting of HRP,.In addition,the HRP-MIP thermalstability tests show that the gel have a good protection in enzyme activity of HRP.The surface of nano-silica gel wrapped Optimization results show that, the use ofγ-MPS modification, with small size (<100nm), and above the phase transitiontemperature of the polymerization temperature were the essential factors for gelformation on the silica surface.This study focused on the imprinting of biological macromolecules and e xploring thefeasibility analysis of the preparation conditions for Imprinted Thermo-sensitiveNanoparticles.
