Perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA), a typical perfluorinated compounds, toxic effects of widespread concern. This study as research subjects in BALB / c mice were fed a normal diet (RD) and high fat diet (HFD), PFOS and PFOA BALB / c mice lipid metabolism through the comparison of the two compounds exposed to different results, looking for its toxic mechanism may provide a scientific basis for the safety assessment of perfluorinated compounds. The PFOS exposure experimental results show: of PFOS exposure can result in BALB / c mice weight loss, reduced food intake; mouse serum triglyceride (TG) and cholesterol (CHOL) levels were significantly decreased high-density lipoprotein cholesterol (HDL-ch affected) and the level of low-density lipoprotein cholesterol (LDL-ch); reduce blood sugar levels in mice as a the blood glucose repository glycogen content decreased; mouse serum thyroid hormones FT3, FT4 reduced the levels of estradiol changes in hormone content; damage liver function, elevated liver fat content biopsy revealed liver cell hydropic degeneration; over-expansion of the endoplasmic reticulum in the liver cells were observed under transmission electron microscopy, and a large number of lipid droplets gather, the liver becomes swollen; peroxisome proliferator-activated receptor PPARa and PPARy expression and fatty acid (FA) the CHOL metabolic key gene expression by a corresponding effect; observed significant difference in results between the mice reared in different feed and different gender also significantly different between the mice. PFOA exposure results show: PFOA is exposed to lower glucose levels in mice can cause BALB / c male mice of weight loss, reduced food intake; serum TG and CHOL levels were significantly decreased HDL-ch and the level of LDL-ch also affected; as a the blood glucose repository glycogen content also reduces muscle glycogen content is unchanged; PFOA exposure of mice after liver MDA content, SOD, GPT and LDH enzyme activity decreased; biopsy revealed hepatocyte ballooning degeneration, liver cells become swollen; ultrastructure expansion of intrahepatic endoplasmic reticulum, mitochondria ridge spreading, but with different PFOS results, lipid droplets of PFOA-exposed mouse liver cells but accumulates in the nucleus; PPARa and PPARy FA, CHOL metabolic expression of key genes are subject to varying degrees of impact. Likewise, these results are obvious differences between mice of different forage. Analyzing the above experimental results, it can be speculated: PFOS and PFOA having a similar structure, with FA can interfere with the FA in the blood and in the transport and metabolism in cells, and interfere with the transport of lipids between the liver and surrounding tissue in the liver by affecting the secretion of LDL , resulting in the accumulation of fat in the liver. PFOS and PFOA different results, suggesting that the two are still the difference in the toxic mechanism, these conclusions need to be confirmed by further studies.
