Abstract


The complete mitochondrial genomes of three species of Crocodylus （Crocodylus acutus, Crocodylus novaeguineae and Crocodylus intermedius） have been sequenced to examine the gene features and arrangements and discuss the phylogeny of crocodylians, especially of Crocodylus. The complete mitochondrial genomes of the three crocodilians are 16, 883bp、16, 894bp and 16, 828bp, respectively. The gene orders of the three species conform to other crocodilians sequenced, which included 22 tRNAs, 2 rRNAs, 13 protein-coding genes, as well as a control region. The difference of gene rearrangement from typical vertebrate can be found in two tRNA genes, i.e. the rearrangement of tRNAPhe, and the tRNASer-tRNAHis-tRNALeu cluster, which showed conservation of the crocodilian gene arrangement.Phylogenetic analyses （maximum likelihood, Bayesian inference） based on the 12 mitochondrial protein-coding genes were performed among 19 crocodilians. We reanalysesed the phylogeny of crocodilians and the interspecies relationships within Crocodylus. The results supported that Gavialis gangeticus join Tomistoma schlegelii on a common branch and then constitute a sister group to traditional Crocodylidae. Crocodylus cataphractus was isolated from Crocodylus and had the closest relations with Osteolaemus tetraspis. In Alligatoridae, Paleosuchus was the sister taxon to Caiman. In Crocodylus, the results agreed this genus to be divided into two separate lineages: New World and Indopacific groups and then C. niloticus form the sister taxon to the New World crocodilians.
