Abstract

Along with the continuous development of modern agriculture, pesticides as an important tool for the prevention and treatment of disease, insects, harm, has gradually become an important part of the agricultural production. However, due to the regulation and the use of some of the holes, the problem of pesticide residues also becoming a shackles of modern agricultural development. Pesticide residues refers due to pesticides stable chemical properties, resulting pesticide after application, can not be completely decomposed, in whole or in part, are left in the surface of the environment or agricultural. These pesticide residues in less than a certain value, not harmful to humans, but it is easy to accumulate in humans and animals, and cause chronic poisoning. In addition, due to the improper application of some agricultural producers over the pursuit of profit, the application of pesticides before harvesting of agricultural products in some of the short production cycle, resulting in serious pesticide residues, pesticide acute poisoning can also cause. Many pesticides, organophosphorus pesticides by agricultural producers sought after because of its low cost, selectivity, and a wide range of prevention, occupy large segments of the pesticide use. But for the charge of people's health as well as the development of green agriculture requirements for detection of pesticide residues in agricultural products into the consumer market is necessary. Which pesticide residues speed the pesticides variety of detection methods, including gas chromatography, high performance liquid chromatography, gas chromatography - mass spectrometry coupled with technology instrument detection method, in addition, there are principles based enzyme inhibition assay measurement methods. These methods have their advantages, vigorously develop applications in different areas. In order to solve the the pesticide residue rapid detection problem, this thesis, described in the following three parts: first prepared the pesticide testing with test strip use the wheat amylase catalytic substrate indophenol acetate reaction environment in pH 6.98 dissolving wheat starch, and then extract the enzyme solution, and reaction was carried out to observe the color change of the reaction liquid. Use a plant esterase detection of malathion pesticide EC, by visual colorimetric determination. A preliminary understanding of the range of reaction conditions required for this reaction system, a pesticide inhibition time, enzyme reaction time, reaction temperature and substrate concentration. 2 introduces a presence in the fast-phase reactions, visual inspection method sensitive methyl parathion. The method of trial a water-soluble materials - polyvinyl alcohol of cholinesterase fixed. When used, will be an enzyme membrane into solution, and the film rapidly dissolving enzyme. Cholinesterase freed from the enzyme membrane sufficiently mixed reaction with the substrate, as well as inhibitors methyl parathion. Clear color change in the same phase system rapidly, compared to more easily distinguish the color difference than the color card, and to overcome the uneven dyeing defect colorimetric card. After inhibition of 10 minutes, 5 minutes of reaction, by visual colorimetric detection to 0.1μg/mL methyl parathion. 3 of the enzyme membrane system is introduced into the nano-particles - hydroxyapatite, and a hydrophilic ionic liquid. Filter out the best ratio of enzyme membrane ionic liquid and hydroxyapatite preparation, and detection of methyl parathion pesticide draw inhibition curve. Select from commercially available lettuce samples to be detected in accordance with the method of operation of the above experiments. In this paper, the above content a relatively new type of enzyme immobilization methods, preliminary preparation of a film speed measured for organophosphate pesticide detection.
