Study on the Inducting Technology and Development-relevant Proteome of Trimolter in Silkworm

The tetramolter can be induced to trimolter by anti-juvenile hormone,the trimoltersilk fiber is fine and uniform and strong tensile properties to reel the high quality finedenier raw silk, it is a rare ideal raw material for develpoment of special purpose rawsilk with high economic value and use value. In order to find related specific proteins toexplain the regulation mechanism of anti-juvenile hormon, we used gel electrophoresisand biological mass spectrometry to study the hemolymp and fat body proteins betweenthe treated groups and control groups based on the induced trimolters. The results asfollow:1. Using polyvoltine silkworm race P50as materials established the inductiontechnology system of trimolters by anti-juvenile hormone.The result showed thatusing400ppm concentration to treat silkworm could obtain the trimolters withshortening instars and enhanced physical, also obtained94percent induction rate of3instar treated groups and96percent induction rate of4instar treated groups withcocoon filament denier of1.224D and0.785D.2. Six different species of silkworms were induced using400ppm based on theinducting technology of trimolter. The results showed that inducing effect anddevelopment of trimolters were closely related to the silkworms varieties and treatedperiod, the inducing effect of bivoltine silkworm hybrids was higher than the polyvotinesilkworm hybrids, Chinese varieties higher than Japanese varities.The cocoon qualitiesof spring silkworm were better than the autumn silkworms’, the finest cocoon fiberwere1.079D and0.803D.We found that Liang guang NO.2, Qiu feng×Bai yu and Baiyu×Qiu feng were suitable for production the trimolters.3. The hemolymph and fat body proteins were ectracted after72hour which waslater of the beginning of the fourth instar with treated and control groups.1DE/MS wereused to separate and identify the hemolymph and fat body proteins,82matchable protein candidates were searched among the hemolymph different bands, by eliminatingthe redundant protein candidates, a total of26protein compositions were identified anddefined as17kinds of proteins or protein subunits, and find that30kD and70kDproteins were rich in treated group hemolymph,at the same lysosomal thiol reductaseIP30isoform1and2were identified, they might be related to the trimolters,diseaseresistance;49matchable protein candidates were searched among the fat body differentbands,12protein compositions were identified, they contained heat shock protein,storage protein and the enzymes and transcription related to gene transcription andtranslation.4. By2DE technology,62protein compositions which differential expression weremore than twice were detected from the fat body proteins between the treated andcontrol group after72hour after the beginning of the fourth instar,21spots whichvolume ratio was more than three times were also detected containing6spots whichwere specific expression.18proteins were identified from21spots which volume ratiowas more than tree times by in-gel digeated and analyzed on MS, contained30kDproteins,90kD heatshock protein, ecdysone oxidase, ecdysteroid-phosphate phosphatase,aldo-keto reductase, BmLSP-T, nicotinic acetylcholine receptor subunit alpha4isoform2precursor and serpin-5.
