Effects of Chlorogenic Acid on Immune Function and Intestinal Barrier Function of Weaning Rats

The objective of this study was to study the effects of chlorogenic acid on growth performance, immune function, and intestinal barrier function in weaned rats. Thirty-two healthy weaned male Sprague Dawley rats (21±1d of age;62.26±2.73g) were selected and randomly allotted to four groups for14days experiments:(1) control weaned rats group;(2) LPS group (lmg/Kg.BW LPS at day13);(3) CHA20group (20mg/kg CHA);(4) CHA50group (50mg/kg CHA). Firstly, we investigated the effects of chlorogenic acid on the growth performance of weaned rats, and then measured the change of physiology and biochemistry, immune function, and intestinal barrier function in weaned rats orally administrated chlorogenic acid.The results showed that the CHA50group had beneficial effect on the growth performance of weaned rats and it significantly reduced the7-14d the ratio of feed:gain (p<0.5). After LPS challenge, the small intestine length index, spleen weight index, serum IgM level, jejunal and ileal villus height and villous height/crypt depth ratio, colon and jejunum occludin and ZO-1protein expression, colon and jejunum tissue IL-10levels were significantly decreased compared to normal group (p<0.05), and the levels of serum IL-2and IFN-y, IFN-y/IL-4ratio, concentration of serum urea nitrogen and albumin, DAO concentration in serum, jejunal and ileal crypt depth, L/M ratio in urine, the content of IFN-y and TNF-a in colon and jejunum tissue were significantly increased (p<0.05). The chlorogenic acid significantly increased (p<0.05) the length of small intestine index, spleen weight index, the content of IgM in serum, jejunal and ileal villus height and the ratio of villous height/crypt depth, the expression of occludin and ZO-1protein in colon and jejunum, the level of IL-10in jejunal tissue and significantly reduced (p<0.05) the levels of IL-2and IFN-y in serum, the ratio of concentration of IFN-y/IL-4, the concentration of urea nitrogen and alanine aminotransferase, jejunal and ileal crypt depth, urine L/M ratio, the concentration of serum DAO, the content of IFN-γ and TNF-α in colon and jejunum tissue. In summary, the CHA treatment improved the growth performance of weaned rats, and alleviated LPS-induced lower immunity, intestinal structure and barrier function injury. It also reduces the intestinal permeability, regulates inflammatory response of intestinal tissue and immune function, and improves gut barrier function.
