Abstract


Objective: This study, aimed to explore the effect of Morinda Officinalis How onadult male (SD) rats’ spermatogenic dysfunction induced by microwave radiation.Methods:Forty adult male SD rats were randomly divided into five groups,namelyblank control group,microwave group,negative control group, aqueous extract ofMorinda Officinalis How group and alcohol extract of Morinda Officinalis How group.The microwave radiation source was a microwave signal generater (900MHZ,1.0W),and the radiation density was218mμ/cm2.All the groups were exposed to microwaveradiation for two weeks except the blank control group,12h/d, then the negativecontrol group was given with double steamed water and the two extract groups weregiven with corresponding extractive of Morinda Officinalis How for twoweeks,once a day.One day fllowing the last dose,the body mass, testis andepididymis were weighted,then the organ index were calculated. At the sametime,the sperm relative number, sperm activate rate and the rate of teratosperm werecalculated.The changes of testicular tissue were observed with optical microscope andelectron microscope.while the ELISA method was used to measure the hormoneleveles of serum T and serum LH.. Besides,the receptor of LH in testicular tissue werelabeled with FITC,then the results were observed with confocalmicroscopy.Meanwhile,the expression leveles of LHR were determined by real-timefluorescent PCR method.The results were shown using the computer image, and the statistics were analyzedwith the SPSS17.0software. Here, the data were illustrated using x±s, and α is set tobe0.05for significant inspection standard.Results:No significant difference on the body mass or on the organ index were foundbetween every two groups.The sperm relative number and sperm activative rate of the groups exposed tomicrowave radiation were significantly lower than the blank control group(p<0.05),however, the rate of teratosperm was significantly higher than theblank control group(p<0.05). Compared with the negative control group,spermrelative number and sperm activative rate of the two extract groups were significantlyhigher(p<0.05). Meanwhile,the rates of teratosperm of the two extract groups weresignificantly lower the negative control group(p<0.05).Compared with the blank control group, the hormone leveles of serum T andserum LH were significantly higher(p<0.05) in the other four groups exposed tomicrowave radiation. The hormone levelels of serum T and serum LH in the twoextract groups were higher than the negative control group(p<0.05). However, nosignificant difference was found between the two extract groups(p＞0.05)..Morphologic changes were found in the results observed with optical microscopeand electron microscope, and the repaired effect of Morinda Officinalis How wasfound in the two extract groups.Compared with the blank control group, the expressions leveles of LHR in theother four groups were significantly lower(p<0.05). Furthermore, the two extractgroups had lower expression leveles for LHR than that of the negative controlgroup(p<0.05).Conclusion:Morinda Officinalis How has the repaired effect on adult male (SD) rats’spermatogenic dysfunction induced by microwave radiation.
