Protective Effect of Pretreatment with Atorvastatin and Pioglitazone on Myocardial Ischemia Reperfusion Injury in Rats

Objective To investigate the effect and mechanism of pretreatment with atorvastatinpioglitazone,atorvastatin plus pioglitazone on myocardial ischemia reperfusion injury in ratsmodel.Methods 54 male wister rats were randomly divided into shame-operated group (n=6,normal saline 2ml/d), I/R group (n=12, normal saline 2 ml/d), atorvastatin group (n=12, atorvastatin 20mg kg-1d-1),pioglitazone group (n=12,pioglitazone 3mg kg-1d-1) and atorvastati+pioglitazone group (n=12, atorvastatin20mg kg-1d-1, pioglitazone 3mg kg-1d-1).After one week cf pretreatment, rats were subjected to 30 minof left-anterior-descending coronary artery occlusion and 120min reperfusion. After reperfusion, nitric oxide(NO), endothelin-1(ET-1),creatin kinase isoenzyme (CK-MB) and lactate dehydrogenase (LDH) in bloodserum were measured. At the end of reperfusion, ischemic and infarct areas were measured by Evans blueand TTC staining respectively..Results①Compared with the control group, the CK-MB, LDH, NO of the I/R group decreased, the ET-1increased.②.Compared with the I/R group, atorvastain, pioglitazone, atorvastain plus pioglitazonedecreased LDH、CK and ET-1, increased the content of NO in blood serum and relieved myocardial ischemiareperfusion injury in rats (all p<0.05).③.Compared with atorvastain and pioglitazone group , Atorvastainplus pioglitazone appeared to have an even better effect than each of single drug group (all p<0.05). Butthere was no significant difference between the two single drug group(all p>0.05).Conclusion Pretreatment with atorvastain, pioglitazone could relieve myocardial ischemia reperfusioninjury in rats, and their joint effect is much more significant, which is partly related by increasing the contentof NO and decreasing the content of ET-1 in blood serum.
