Abstract


Background: Hepatocyte cell adhesion molecule (hepaCAM) isfound by Shen S in2004. The protein is typical of a transmembraneadhesion molecule. Exosomes are vesicles surrounded by a phospholipidbilayer (approximately30–100nm in diameter), secreted by many kinds ofcells. It includes many kinds of protein and RNA. There are many kinds ofproteins present in cancer cells. These proteins have many functions.Tumor cells could secrete proteins such as HSP70, HSP60throughexosomes. The presence of hepaCAM outside the cells and its function hasnot yet been reported.Objective: This study was to investigate that whether hepatocyte celladhesion molecule (hepaCAM) is actively secreted from renal cell cancertransfected with hepaCAM by a pathway involving exosomes and whetherhepaCAM could inhibit the proliferation of renal cell cancer.Materials and methods: a recombinant adenovirus vectors plasmidcontaining hepaCAM was transfected into786-0cells. The786-0cellstransfected with empty adenovirus vectors plasmid and untreated786-0 cells were as control.The expression of hepaCAM was detected by westernblot. The transfected cells was treated with monensin and5-(N-N-Dimetyl)-amiloride hydrochloride (DMA), and then extracellularculture media and exosomes were used to investigate the present ofhepaCAM.Results: HepaCAM adenovirus vectors plasmid was successfullyconstructed and stably transfected into786-0cells. Monensin enhancerelease of hepaCAM and DMA inhibit release of hepaCAM fromtransfected786-0cell. HpeaCAM is found in exosomes by western blot.HepaCAM inhibit the proliferation of786-0cells by MTT.Conclusions: HepaCAM is successfully transfected into786-0cells.HepaCAM is secreted from renal cell cancer by a pathway involvingexosomes. Exosomes collected from786-0cells transfected withhepaCAM adenovirus plasmid inhibit proliferation of renal cell cancer.
