Abstract


Chapter1Establishment of rat model of epirubicin induced premature ovarian failureObjective Observation of epirubicin (EPI) effect on the number of follicles in ovarian and sex hormone level in rats, to establish animal model of epirubicin induced ovarian damage in rats.Method Female Wistar rats the research object, to the standards of the24rats were randomly divided into3groups,8rats in each group, For Control group, EPI15mg/kg group, EPI30mg/kg group. At the beginning of first, respectively, in different treatment. Control group without treatment, EPI group15mg/kg, EPI15mg/kg,30mg/kg EPI30mg/kg solution group rat tail vein injection of different doses of,8days at a time. The general conditions of rats after administration, including rats, eating, body weight and the estrous cycle, All the rats were respectively before administration, after administration of2D4D,6D,8D,10d,12D via tail vein blood1ml measured estradiol, follicle-stimulating hormone, For all rats were killed12days after administration, the ovarian HE staining for paraffin sections, to observe the changes of morphology and the number of follicles in ovarian.Result After administration of the EPI treatment group weight were decreased in different degree,appetite than the previous administration decreased significantly and dose and time dependent (F=174.198,226.294,282.294, P=0.000). After treatment, Control group estrous cycle still rules:EPI15mg/kg rat estrous cycle, although complete, but was extended to8-9days, EPI30mg/kg rat estrous cycle. After treatment, Control group of estrous cycle still rules; EPI of rats in the15mg/kg group has complete estrous cycle, but the extension to8-9days. EPI of rats in the15mg/kg group has complete estrous cycle, but the extension to8-9days. EPI30mg/kg group rats with estrous cycle dysfunction, long time stagnation in diestrus. EPI treatment decreased the level of E2group rats, the elevated levels of FSH, two EPI FSH treatment group and Control group, there were significant differences in E2levels between the groups was statistically significant (F=7.593,10.405p=0.004,0.001).EPI30mg/kg group than in EPI group15mg/kg E2level decreased more significantly elevated levels of FSH is more obvious, difference between the two groups was statistically significant (p<0.05).12th days after the administration of HE staining was observed in EPI group compared with Control group, the number of follicles was significantly reduced, significant differences between the groups, with statistical significance (F=381.360,50,258,204.646,54.837p=0.000).EPI30mg/kg group than in15mg/kg group EPI number of follicles built more obvious, significant differences between the groups, statistically (p<0.05).Conclusion After the treatment of EPI rats decreased body weight, decreased appetite, negative feedback significantly increased the FSH level decreased by E2, Estrous cycle of rats disappeared, and the number of follicles reduced resulting in impaired ovarian function. Application of30mg/kg successfully established an animal model of premature ovarian failure, can be used as the research foundation for the next step.Chapter2The effecti of DHEA on epirubicin induced ovarian damage in ratsObjective To investigate the protective effect of DHEA on EPI induced ovarian injury.Method Female Wistar rats as the research object, the32rats were randomly divided with the experimental criteria into4groups,8rats in each group, Contrl group, EPI group, DHEA group, EPI+DHEA group. So the first day were given different treatment:no treatment, intravenous injection of EPI30mg/kg, DHEA5mg/kg gavage, EPI30mg/kg tail vein injection, DHEA5mg/kg gavage+EPI30mg/kg tail vein injection. Experiment8days at a time. The general conditions of rats after administration, including rats, eating, body weight and the estrous cycle, All rats were killed12days after administration, the ovarian parallel paraffin, HE staining, to observe the changes of morphology and the number of follicles in ovarian.Result Body weight of rats in the Control group with time due to increased, coat gloss, limbs activity and diet size had no significant change. Body weight of rats in the EPI group after administration into different degrees of decline. Diet than the previous administration decreased, limb weakness, decreased activity, hair dull dull, size reduction. Body weight of rats in the DHEA group with time due to increased, coat gloss, limbs activity and diet size had no significant change. The weight of EPI+DHEA rats decreased more slowly, after administration of sixth body weight with time and growth. EPI group compared with EPI+DHEA Group4d,6d,8d,10d,12days and two significant differences between the groups, with statistical significance (P<0.01).4d,6d,8d,10d,12days among the three groups had significant difference weight(P<0.01). After administration of microscope in group EPI, group EPI+DHEA total follicles, growing follicles, the number of mature follicles were less than group Control, Group DHEA total follicles, primordial follicle, mature follicle counts more than the Control group, EPI group and Control group were significantly different (P<0.01); There were significant differences in EPI group and EPI+DHEA group the number of follicles (P<0.01).Conclusion DHEA promotes ovarian primordial follicles to growth process, improve the EPI causes the ovarian reserve function.
