The Application of STK1and AFP in Therapeutic Monitoring of PLC Pre and Post TACE

Objective:The research aims to examine the expression levels of STK1and AFP pre and post TACEamong PLC patients, to analyze the correlation and efficacy, to explore its clinical significance,and to provide theoretical basis for the PLC treatment monitoring.Methods:50subjects with PLC in experimental group and51cases in healthy control group wererandomly chosen. Experimental group was followed up2weeks before TACE,4,8,12,16weeksafter TACE, while the control group was followed up2weeks before TACE and4weeks,8weeks after TACE. Western blotting-enhanced chemiluminescence was used to examine TK1level, electrochemiluminescence immunossay is used to examine AFP density. Data wereanalyzed with SPSS13.0, including χ~2test, t test and relative analysis.Results:1. In the PLC without metastasis group, TK1density rose after the operation (0.76±4.91)and decreased after8,12,16weeks(3.7±3.45,3.69±3.56,3.59±3.50). AFP density decreasedafter4,8weeks (3.7±7.11,2.00±5.50), and it rose after12weeks and16weeks(0.93±3.14,1.83±3.86);2. In the PLC with metastasis group, TK1density(pmol/l) tested4times after the operationrose, comparing the density two weeks before the operation (2.92±5.60,3.42±4.37,2.50±4.68,4.17±5.30);3. In the PLC death group, TK1density rose after4weeks, and reached the highest in thetwelfth week, while the AFP density had no variation. In the lost follow-up group, TK1densityrose4weeks after the operation and decreased8weeks after the operation.The variation of AFPdensity is not obvious;4. There was difference among three groups in TK1in T0, T1,T2(between the groups,F+14.35, P<0.05; interclass, F=13.20,P<0.05) while there in no difference in AFP(betweenthe groups,F=0.64,P>0.05;interclass,F=1.296,P>0.05);5.There was significant difference between the experimental and control group in TK1andAFP density which both rose before the operation,(P<0.01); 6.38cases were followed up12weeks after TACE. Comparing the PLC with metastasispre and post operation, there is significant difference in TK1density and AFP density whichboth rose. The comparison of PLC without metastasis pre and post operation shows TK1density decreased and the difference is significant (P<0.05), while AFP density decreasedwithout significant difference (P>0.05). There is significant difference in TK1density betweenPLC with metastasis and PLC without metastasis (P<0.05), while there is no difference in AFPdensity (P>0.05);7. PLC without metastasis have good iodized oil accumulation and TK1density decreasedgreatly and had great variation. AFP density has some changes but not big. PLC with metastasisdidn’t have good iodized oil accumulation and there are no big changes in TK1and AFPdensity.Conclusion:1. High expression of TK1and AFP in PLC serum indicates that TK1is similar to AFP inthe application of PLC diagnosis.2. TK1density is high in PLC with metastasis group and low in PLC without metastasisgroup, while AFP density is high in both groups which indicates TK1serves better in clinicalmonitoring and can be used as an biological monitoring index to provide clinical references
