Abstract


[Objectives] By using the electrospinning technique and biomimetic mineralization technique comprehensively, it is aim to study the mineralization characteristics of Gelatin(Gel)/Polycaprolactone(PCL) composite fiberous membrane in vitro,and study various influence factors of nucleation and mineralization, in order to manufacture the ideal mineralization composite fiberous membrane and explore the new bone tissue engineering scaffolds.[Methods] Firstly, Gel/PCL composite fiberous membrane, HA/Gel/PCL composite fiberous membrane, and CaCl2/Gel/PCL composite fiberous membrane in different concentrations are obtained by the electrospinning technique. All of the samples are analyzed with Scanning electron microscope-Energy dispersive spectroscopy (SEM-EDX), Fourier Transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD). Secondly, for accelerating the minerization process, all of the samples are immersed in ten times simulated body fluid (SBF) for1,2, and4hours respectively, all of the analysis metioned above are redone to know the influence of composite fiberous membrane with different concentrations of hydroxyapatite or calcium chloride.[Resluts](1) With the increasing content of Gel from0%,2%,5%to8%, the composite fibers become smoother and more even, and the diameter of the fibers increase accordingly.(2) HA can be electspinned into the composite fiber of5%Gel/8%PCL. With the increasing content of HA from1%,1.5%to2%, the diameter of the fibers decrease accordingly. (3) Calcium chloride can be electspinned into the composite fiber of5%Gel/8%PCL With the increasing content of calcium chloride from1%,1.5%to2%, the diameter of the main fibers decrease accordingly. Meanwhile, lots of finer netlike structure can be seen between the main fibers, which is positively correlated with the content of calcium chloride. The finer fibers have obvious smaller diameter than the main fibers, which leads to bigger specific surface area.(4) Compared with Gel/PCL composite fiberous membrane, with the increasing content of HA from1%,1.5%to2%, the mineralization speed increases accordingly in ten times SBF within4hours.(5) Compared with Gel/PCL composite fiberous membrane, with the increasing content of calcium chloride from1%,1.5%to2%, the mineralization speed increases accordingly in ten times SBF within4hours.(6) Compared with Gel/PCL composite fiberous membrane, the morelike structure of bone trabecula can be seen in the mineralized composite fiberous membrane within HA or calcium chloride.[Conclusion] The electrospinning composite fiberous membrane obtained in this study has the molecular structure of amino group, carbonyl group and apatite, similar to the extracellular matrix. In vitro, it can be manufactured by biomimetic mineralization methods and takes the shape of apitite-like structure.
