Effects of Nonylphenol and Tributyltin on Immune Factors, Vitellogenin Gene Expression and Gonad Development in Macrobrachium Rosenbergii

Abstract


The effects of different doses nonylphenol (NP) and tributyltin (TBT) (0.1mg/L, 0.2mg/L, 0.4mg/L) on immune factors, vitellogenin gene expression and gonad development of Macrobrachium rosenbergii were researched with the molecular biology, the histology and the micro technology. The main results as followings:1 Effects of NP and TBT on SOD, ACP and AKP activities in Macrobrachium rosenbergiiThe M. rosenbergii was exposed to NP at different doses within 48h. The activity of superoxide dismutase (SOD) in serum at the high dose group was significantly inhibited (P <0.05) in the early stage, the activities of SOD in serum were increased at each dose groups after 24h, and at medium and high dose groups were significantly different(P<0.05) from the control after 24h. The activities of Acid Phosphates (ACP) in serum, which were decreased at medium and low dose groups, was significantly lower (P <0.05) at the low dose group within 12h, but at the high dose group showed induced and then inhibited. The activities of Alkaline Phosphatase (AKP) in serum were inhibited at first and then induced at each dose groups, the activities of AKP in serum were significantly higher (P<0.05) at the medium and low doses compared with the control within 48h. The M. rosenbergii was exposed to TBT at different doses within 48h. The activities of SOD in serum were significantly inhibited (P<0.05) at the low dose group within 12h, and were increased after 48h. The activities of ACP in serum were significantly inhibited (P<0.05) at medium and low dose groups within 24h, while the activities of ACP in serum were decreased transiently and then increased at the high dose group, and the activities of ACP in serum was significantly higher (P<0.05) compared with the control after 48h. The activities of AKP in serum were significantly decreased in the early stage (P<0.05) at medium and high dose groups, but the activity of AKP in serum was significantly increased (P<0.05) at the low dose group. The activities of AKP in serum were increased at each dose groups after 48h, and at medium and high dose groups were significantly different from the control group (P<0.05).2 Effects of NP and TBT on VTG gene expression in Macrobrachium rosenbergiiExpression of vitellogenin (VTG) at exposure in NP was observed increase in hepatopancreas and ovarian of female M. rosenbergii, induction effect of low dose group were stronger than medium and high dose groups. Expression of VTG gene of testis and hepatopancreas in male M.rosenbergii was also promoted at exposure in NP, of which dose effect relationships existed in hepatopancreas, induction effect were more obviously with the doses increased, but significantly higher expression of VTG in the testis was only observed in the low and medium dose groups. Expression of VTG increased in the hepatopancreas of female M. rosenbergii treated by TBT, however, which decreased in ovarian. Expression of VTG in hepatopancreas of male M. rosenbergii were inhibited by TBT, however, effect of TBT on expression of VTG in testis was non-significant.3 Effects of NP and TBT on gonad development in Macrobrachium rosenbergiiDevelopment of testis and ovary in M.rosenbergii was inhibited at exposure in NP, inhibition of testicular and ovarian development gradually increased with the exposed time extending, in which the dose-time response relationship was obvious. Testis development of could be induced at exposure in TBT, and with exposure time prolonged, development of testis was accelerated gradually, but effect on ovarian of M.rosenbergii was non-significant
