DDP and Hyperthermia for Intraoperative Autotransfusion in Liver Cancer Surgery: Elimination of Carcinoma Cells and Effect to Erythrocytes

Objective To investigate the role of DDP and hyperthermia for intraoperative autotransfusion to eliminate carcinoma cells and the effect to erythrocytes in liver cancer surgery.Methods Cultured hepatoma carcinoma cells were mixed with erythrocytes suspensions and separated to 6 groups by temperature and DDP concentration, A: 37℃and DDP 0μg/ml, B:37℃and DDP 100μg/ml, C:37℃and DDP 200μg/ml, D: 42℃and DDP 0μg/ml, E:42℃and DDP 100μg/ml, F:42℃and DDP 200μg/ml, then respectively treated for 30 min. After that, tumer cells were isolated from RBCs using density grandient centrifugation. Part of the isolated tumer cells were tested for inhibition ratio by MTT method. The rest tumer cells were cultured and their clone formations were checked. The isolated erythrocytes suspensions were tested for osmotic fragility and 2,3-DPG.Results The effect of eliminating carcinoma cells in a DDP centrifugation and temperature dependent manner (P<0.01). The inhibition ratio of tumer cells is more than 99% and there is no clone formation can be tested in group F. The osmotic fragility of RBCs is slightly modified in group F, however the hemolysis ratio is no more than 1% when the sodium chloride is 0.68%. There is no deference of the RBCs’ 2,3-DPG between before and after being treated in group A and group F (P<0.01)Conclusion When treated at 42℃and DDP 200μg/ml for 30 min, all hepatoma carcinoma cells in the recycling blood lost proliferation ability and there is no effect to the transport oxygen ability of the erythrocytes.
