Objective:To study the inhibitive effect of Resveratrol (Res) on human cervical cancer HeLa cells in vitro and its mechanism.Methods:The HeLa cells were treated by different concentration of Res, the concentration were as follow:17.5、35、70、140μmol/L. The viability of HeLa cells were measured by MTT assay. The HeLa cells were treated by 70μmol/L Res for 24h. Expression of phosphorylated eykaryotic initiation factor 4E(p-eIF4E)、phosphorylated 4E-binding protein 1(p-4E-BP1) and cysteine aspartic acid specific protease 3(caspase-3) were determined by Western Blot analysis.Results:The HeLa cells were treated by different concentration of Res on different time, and there was a significantly inhibitive effect on Hela cells, with dose and time—related effect. At the same time, with the increasing of concentration of Res, the ratio of proliferative inhibition of Hela cells was increasing(F=93.76、637.22、270.23, P<0.01); At the same concentration, following the lasting of time, the ratio of proliferative inhibition of Hela cells was increasing(F=29.99、12.88、8.79、9.42, P<0.01). With the HeLa cells treated by 70μmol/L Res for 24h, the expression of p-eIF4E and p-4E-BPl was significantly lower than those in the control group(T=14.93、4.78, P<0.01)and the expression of caspase-3 was significantly higher than those in the control group(T=10.68, P<0.01)..Conclusion:This study indicated that Res had significantly inhibitive effect on the HeLa cells in vitro. It probebaly correlated with the down-regulating of p-eIF4E and p-4E-BP1 and activation of caspase-3, and then causing apoptosis of the HeLa cells.
