The Effects of Pretreatment on the Germination and Salt Resisitance in Switchgrass

Bioenergy crop, switchgrass was usually grown in the marginal land to avoidcompetition with food crops. The growth of early stage of switchgrass can influence the finalyield. Many seed treatment methods were used to improve the germination and seedlinggrowth of switchgrass in the previous study, however no references reported the effects ofultrasound on switchgrass seed,and few literature can be found about the effects of seedtreatments on germination and seedling growth under salt stress.In our study, using modelling analysis, conditions for the ultrasound treatment ofswitchgrass seeds,including sonication time (factor A), sonication temperature (factor B) andultrasound output power (factor C), were optimised for stimulating seed vigour of switchgrassseeds using an orthogonal matrix design [L16(45)]. Additionally, water uptake tests with threetreatments(control, soaking in water only and then redried and ultrasound treatment and thenredried) were performed to investigate further details of the mechanisms for stimulatinggermination using ultrasound.The combined pretreatments including ultrasound irradiation and KNO3soaking wereused to overcome salt stress during germination and seedling growth. In addition,9indicatorsincluding3physiological indicators were investigated. Model analysis and principalcomponents analysis (PCA) were used to integrate the indicators and build the interactionbetween factors and indicators.We found that:1.The final optimal ultrasonic treatment conditions were found to be a sonication time of25min, sonication temperature of38℃and output power of300W.2.Both water and ultrasound per se play important roles in increasing water uptake at theearly stage of soaking.3.The combined pretreatments were effective in terms of overcoming the salt stress ofswitchgrass seed.4.The important extents of the four treatment factors were time of KNO3treatment>sonication temperature>sonication time> concentration of KNO3under salt stress.5.A network linking the indicators and factors was established.
