Biodegradable amphiphilic block copolymers, due to its good biocompatibility and biodegradability, is widely used in the biomedical field, including drug / gene delivery systems, tissue engineering. In this paper, the ring-opening polymerization (ROP) and atom transfer radical polymerization (ATRP) method of combining design and synthesis of two pH-responsive amphiphilic triblock copolymer, study their self-assembly behavior in aqueous solution, and preliminary their potential application in drug delivery and gene delivery. This paper reads as follows: the first part of the amphiphilic triblock copolymer mPEG-b-PCL-b-PDMAEMA Sn (Oct) 2 as a catalyst, polyethylene glycol monomethyl ether (mPEG) as the initiator the ring-opening polymerization of synthetic diblock copolymers of ε-caprolactone (ε-CL) of mPEG-B-PCL; then with α-bromo-isobutyryl bromide ATRP macromolecular initiator agent of mPEG-b-PCL- Br, and then by ATRP method initiator methacrylate -2 - (N, N-dimethylamino) ethyl methacrylate (DMAEMA) polymerization, to obtain an amphiphilic triblock copolymer MPEG-b-PCL-b-PDMAEMA. By FT-IR, 1H NMR, GPC, TGA on the chemical structure of the polymer, the composition, molecular weight and molecular weight distribution were characterized. The use of fluorescent probes to the TEM and HPPS study amphiphilic block copolymer self-assembly behavior in different pH buffer. The results show that the aqueous solution of the copolymer acts by affinity the water chain segment length, the pH value of the aqueous phase factors. Stability of the polymer Experimental results show that the amphiphilic block copolymer is likely to occur under acidic conditions, the rupture of the ester bond is hydrolyzed, and the polymer is relatively stable under neutral conditions. Cytotoxicity test (MTS method) The results indicate that the toxicity was significantly less than at the same concentration of the tri-block copolymer of branched polyethyleneimine (25 kDa), having a good cell compatibility. In addition, we studied the role of the copolymer with DNA, and for the in vitro release of the hydrophobic drug naproxen. Experimental results show that the ability of the polymer to bind DNA with PDMAEMA segment growth enhancement;, the drug loading of the polymer increases, the rate of drug release decreases with the growth of the PCL hydrophobic segment; under alkaline conditions The drug release rate is faster than under neutral conditions. The second portion of amphiphilic triblock copolymer PEEP was-b-PCL-b-PDMAEMA first synthesized bifunctional the initiator agent 2 - hydroxyethyl-2 - bromo-isobutyrate (HEBI) and cyclic monomer 2 - ethoxy-2 - oxo-1, 3, 2 - dioxo-phospha-cyclopentane (EEP). Sn (Oct) 2 as catalyst, with HEBI ring-opening polymerization of ε-CL HEBI-PCL homopolymer, and then use the terminal hydroxyl groups in the HEBI-PCL further to the ring-opening polymerization of EEP give PEEP-b-PCL two block copolymer. Finally, the polymerization initiator DMAEMA by ATRP method obtained amphiphilic triblock copolymer PEEP was-b-PCL-b-PDMAEMA. 1H NMR, GPC chemical structure, molecular weight and molecular weight of the polymer distribution characterization, the experimental results show that, HEBI-PCL homopolymer and PEEP-b-PCL diblock copolymers have been successfully synthesized.
