Abstract


Cinnamomum kanehirae Hay belongs to the Lauraceae Cinnamomum plant, black cinnamomum camphora again, for Taiwan’s unique evergreen broad-leaved large trees, trunk straight and tall, the primary leaf color changes, deep high ornamental value. And there were great potential for this landscape tree in Fujian. Antrodia Camphorata, Antrodia Cinnamonea, also known Antrodia camphorate, Camphor in the mushroom, Red camphor, etc, only grew up in the hollow trunk of Taiwan unique Cinnamomum kanehirae trees, as a kind of wood decay fungi, and it also is a precious medicinal herbs with extremely high medicinal value. Because of Cinnamomum kanehirae Hay’s unique to Taiwan tree species, this paper were studying on its vegetative propagation technology including cutting, tissue culture and grafting, to establish high cutting survival rate technology system, tissue culture seedling optimization technology system and grafting efficiently survival system. And using these systems production and promotion Cinnamomum kanehirae Hay on a large scale to increase species diversity in Fujian area, also to provide an excellent greening plants for landscape. At the same time, Taiwan national treasure Cinnamomum kanehirae Hay will be extended to mainland to achieve resource share.1.The study on the cutting propagation technology of Cinnamomum kanehirae HayThe research results on the Cinnamomum kanehirae Hay cutting slips of different plant growth regulator (NAA, IBA and ABT1) as well as concentration level processing showed that:The rooting effect of treatment6with deal with1s by2000mg·L-1IBA was most apparent, rooting rate87.0%; While the rooting effect of treatment9with deal with5s by2000mg·L-1IBA was the least effective, the rooting rate only35%. The comparison of the effect of plant growth regulator on rooting rate was in order:IBA>NAA>ABT,.The result of three different cutting types (with crown main branch, with crown lateral branch, no crown lateral branch) treated with2000mg·L-1IBA indicated that:Crown cuttings compared with no crown cuttings, crown cuttings were competitive in rooting rate and root growth situation; While the comparison in basal necrosis rate, the crown lateral branch was minimum, followed by crown main branch, the highest for no crown lateral branch. Thus, with crown lateral branch were better than the rest of the two in both rooting rate and survival rate, and no crown lateral branch had the lowest rooting rate as well as survival rate.The Cinnamomum kanehirae Hay cutting slips had been firstly treated three ways of mechanical damage including cutting leather on both sides, longitudinal split and chafing at base and then treatment by2000mg·L-1IBA before they were planted in matrix. And then these cutting seedlings were cultivated in the automatic spray greenhouse. The results showed that cutting leather on both sides and longitudinal split rooting rate was up6-9%from control group, while chafing at base compared with control group only increased by1%. Consequently, the rooting rate of cutting seedlings that had been deal with the trauma can be improved in different extent. But the results also indicated that after dealing with the three types of trauma, the base part of stem of cuttings necrosis rate would increase by about3-8%, especially cutting leather on both sides, and no matter what trauma treatment all failed to well improve the cuttings rooting number. So that, Cinnamomum kanehirae Hay cutting slips do not need to take special way of trauma treatment, because these treatments not only lower the quality of roots but also increase operation cost.2. Study on the tissue culture technology of Cinnamomum kanehirae HayIn the induction time, the results showed that different concentrations plant growth regulator6-BA, NAA and IBA all could induce the budding, but the treatment of WPM+6-BA1.5mg·L-1+IBA0.1mg·L-1(Hy7) induced the highest number of buds and the bud seedlings grown best. The analysis of range and variance for bud germination and the number of bud proliferation came to a conclusion that:the primary and secondary relations of the impact of various factors on Cinnamomum kanehirae Hay was6-BA>IBA>NAA.In the proliferation of bud, the results of using different species and concentrations of plant growth regulator (6-BA、IBA and NAA) showed that with the increased concentration of cytokinin6-BA, buds multiplication factors increase, and with the treatment of WPM+6-BA2.5mg·L-1+IBA0.25mg·L-1(Nj7) got the highest monthly average buds multiplication factors, up to2.8. After the Cinnamomum kanehirae Hay explant had been subcultured multiplication for30days, taken the range analysis of monthly average buds multiplication factors and found that the primary and secondary relations of factors6-BA> IBA> NAA.In the strong seedling culture, there were mainly to add a certain concentration of cytokinin6-BA,vitamin VB1and exogenous additives-peptone. The results showed that the treatment of Nz5(WPM+6-BA1.0mg·L-1+VB10.1mg·L-1+peptone1.5g·L-1) could not only make buds multiplication factor high, but also could make bud seedlings growing strong and leaf pale green. At the same time, the analysis of range and variance on the monthly average buds multiplication factors of the seedlings cultured30days found that the impact factor on the result of cultivation of size6-BA>peptone>VB1.In the root culture, the main research was that concentration ratio of the two kinds of plant hormone including auxin IBA, NAA and cytokinin KT took the impact on the root culture of Cinnamomum kanehirae Hay. The experiment results showed that the Ng4(WPM+IBA1.0mg·L-1+KT0.1mg·L-1) and Ng5(WPM+IBA1.0mg·L-1+NAA0.5mg·L-1+KT0.15mg·L-1) could get a higher rooting rate (86.7%and80.0%) and number of roots (8.87and8.20, and they also could make the seedlings maintain good and the good root growth potential in the process of root culture. The primary and secondary relations of the various factors influence on rooting rate was that:IBA>KT>NAA.In the seedlings acclimatization and transplant, the tissue culture seedlings with robust buds, grown well, developed root system and plant height of about above2cm had been refined for a week (that is, firstly refining plants4days in closed bottle and then3days in open bottle), and they were transplanted in the outdoor sterile matrix (vermiculite and peat1:1). After that, every other week they would been sprayed once fungicide and watered once most of the tree. After a month, the survival rate of statistics found that the transplanting survival rate of Cinnamomum kanehirae Hay could reach75%and the seedlings grown well, leaf green. But some transplanted seedlings appeared browning death phenomenon in succession. As a result, this also is the key of the Cinnamomum kanehirae Hay studies in future.3.Study on grafting technology of Cinnamomum kanehirae HayThe grafting experiment of Cinnamomum kanehirae Hay found that if the grafting plants could survive, their grafting junction would heal well (figure3-1B and figure3-2B).The research on the effect of different grafting methods of Cinnamomum kanehirae Hay on survival situation showed that the grafting survival ratio with using cut grafting was higher than using cleft grafting. In December2011taking the first survey of grafting survival ratio for the plants grafted in November, the survey data exhibited that cleft grafting method was86.7%, cut grafting method93.3%, with skin grafting method53.3%, side grafting method80.0%, and abdomen grafting method80.0%. But in May2012,that is through a half year after the grafting, taking the secondly survey of grafting survival ratio found that the survival ratio significantly decreased, the cut grafting33.3%, skin grafting and side grafting both13.3%, cleft grafting20%, abdomen grafting only6.7%.The research on the effect of cuttings length of Cinnamomum kanehirae Hay on survival situation showed that:After six months to observe the grafting seedlings survival rate and grafting seedlings height, the grafting survival ratio of9cm cuttings was46.7%,3cm and6cm cuttings both33.3%; In terms of grafting growth potential, through surveying the seedling plant height found that6cm cuttings (plant height36.8cm) grown and developed best, leaf green color and blade thick, while3cm and9cm cuttings (plant height were26.2cm and32.6cm) were not as good as6cm cuttings (see table3-18and figure3-2,3-3).The research on the effect of graft height on the grafting seedlings survival ratio showed that:After a month to observe, graft height of10cm got the highest survival rate80%, while the survival rate of graft height of65cm was the lowest, only26.7%, graft height of15cm was60%,25cm was73.3%,45cm was66.7%; After six months to observe again, graft height of10cm still got the higher survival rate66.7%, while the survival rate of graft height of65cm was the lowest,only6.7%, graft height of15cm was33.3%,25cm was46.7%,45cm was40.0%.(see table3-19and figure3-4).
