Abstract


Objective: To investigate the effects of high molybdenum (Mo) on morphologicchanges and Spermatogenic function of small-tail-fat-Sheep testis.Methods: Total of twelve two months kids were randomly divided into there groupsand feed on diets as follows: control diet (groupⅠ) and high Mo diets (Mo30mg/kg,group Ⅱ;60mg/kg, groupⅢ) for120days. At the end of the treatment, testes andepididymis were examined to study the effects of high doses of molybdenum on thereproductive function on the level of microscopic, microstructural, cell, protein and soon, by the transmission electron microscope, hematoxylin-eosin staining, kit,immunohistochemistry and other technique means.Results:1. Sheep in30mg/kg and60mg/kg groups indicated the body weight restrainedfrom the ninth week (P<0.01). The testis index were significantly decreased in bothgroup Ⅱa ndⅢ (P<0.01) were than that in groupⅠ (P<0.05).2. Compared with group Ⅰ, in all test groups, the rate of deformity increased, spermmotility and density depressed were extremely remarkable (P<0.01).3. Under electron microscope, mitochondrion edema and endoplasm reticulum dilationwere almost everywhere in both groupⅡandⅢ. In addition, vacuoIized mitochondria,increase in apoptosis of spermatogonia, acrosomal defect and teratosperm were found inexposed groups, more over, the higher the dose the more serious damage.4. The statistical analyses by FCM indicated that the G0/G1phase was increased and theG2+M phase, S phase and the PI were decreased in high Mo group (P<0.01).5. Compared with the control group, in all high Mo groups, the content of MDA andTNF-a increased, SOD and GSH-Px depressed were extremely remarkable (P<0.01).Conclusion: High doses of molybdenum (30、60mg/kg) destroyed the normal structureof testis tissue and have effects on sperm quality and reproductive function.
