Abstract


Purpose:To observe the effects on systolic blood pressure, myocardial collagen andMMP-2, MMP-9, TGF-β1of spontaneously hypertensive rats (SHR) withelectro-acupuncture at Zu San Li, Qu ChiTo study the improvement situation of hypertension with the treatment ofelectro-acupunctureTo explore the improvement mechanism of myocardial fibrosis with thetreatment of electro-acupunctureMaterial and method:30SHR rats randomly divided into three groups with the method of randomizedcontrol:1. SHR group (given distilled water)2. Captopril group (given captopril0.4mg.kg-1d-1)3. Acupuncture treatment group (acupuncture at Zu San Li, Qu Chi).10age-matched and normal blood pressured Kyoto rats (WKY) were grouped as WKYgroup (given distilled water).Using BP-98A, the intelligent non-invasive bloodpressure meter, to test the change of systolic blood pressure on caudal artery;After six weeks treatment, take myocardial tissue of each rat, microscope observeHE dyed myocardial tissue biopsy and the content of immunohistochemical staineddetecting collagen component (type I and type III collagen), with RT-PCRdetecting method to detect the expression of matrix metalloproteinase (MMP-2,MMP-9), transformation growth factor-β1(TGF-β1) mRNA.Results:Compare with SHR group (207+16.7mmHg), the results of systolic bloodpressure on captopril group and acupuncture group are165.8±10.2mmHg, and168+13.5mmHg. The results indicate that after treatment, the systolic bloodpressure of these two groups of rats has been changed significantly (P <0.05); Comparing with WKY group, SHR group appears obvious interstitial cellproliferation, and because of compression atrophy, myocardial cells appearvacuolar degeneration, Myocardial fibers toward disorder. Comparing with SHRgroup, the myocardial cells of captopril group, and acupuncture group are veryneat, and their myocardial interstitial fiber are to tend to order; Comparedwith WKY group, SHR group appears obviously in a significant increase in thenumber of cell collagen and myocardial interstitial fibers, the myocardialinterstitial fibers are coarsened and toward disorder,Comparing with SHR group,the myocardial interstitial fibers of captopril group, and acupuncture groupare fine and uniform; Comparing with WKY group, the expression levels ofmyocardial MMP-2, MM P–9is significant reduced, and the expression levelof myocardial TGF-β1is significant increased (P <0.01). Comparing with thecaptopril group and acupuncture group, the expression level of myocardial MMP-2, MMP-9in SHR group is significant increased, and the expression levelof myocardial TGF-β1in SHR group is significant reduced (P <0.05).Conclusion:There is a better antihypertensive effect on SHR rats when we acupuncture atZu San Li, Qu Chi; The acupuncture treatment effectively improves myocardialfibrosis; and protects the myocardial system.
