Abstract


Purpose:ThisstudyintendstoobserveAntaioralliquidonmiceimmune-relatedcytokines, explore its immune mechanism.Material and method:1Material:1Material:Health BALBI/C mice, Male and female,Weighing18-22g、S180tumor strain、H22tumor strain, Cyclophosphamide, heparin sodium injection,automatic microplate reader, BC3000automatic blood cell analyzer, MODEL DS-671electronic scale models、Murine interleukin-2ELISA kit, mouseinterleukin-4ELISA kit, mouse INF-γ ELISA kit.;Drugs: Aetna Oral: guzhi、yiyiren、baibiandou、shaoyao、zhigancao、huangqi、banzhilian Etc. Zhenqifuzhengparticles.2. Method:2.1Modeling:2.1.1Establish the tumor models: take those have been vaccinated7days andin good spirits,full Ascites tumor mice,With saline at an appropriatedilution ratio, Counting the number of tumor cells, And adjust the cell numberto1×107个/ml. before Tumor strain inoculation, Both with0.2%trypan bluestaining, count Live cell rate>95%. Mice were injected subcutaneously in theright front armpit0.2ml S180cell suspension (approximately2×106cells),make the solid tumor model. Except the blank group, the other groups are tobe modeling, a total of40mice modeling, male and female, are at24h aftermodeling starting gavage for consecutive10days, in which each experimentalgroup were given appropriate medication, model control group and the controlgroup were given equal doses of0.9%saline.2.2Animal groups::BALBI/C mice were randomly divided into five groups of10, male and female. Were as follows: control group, model control group, groupZhenqifuzheng particles, Aetna oral low-dose group, ING oral high dose group.In addition to the control group, and the remaining four groups weretumor-bearing mice.2.3Animals administration: Mice in each group daily intraperitoneal injectionof cyclophosphamide20mg/kg, experimental groups gavage0.4ml/times (quiteAetna Health oral low dose prescription drug1.13g, Aetna Health oral high doseprescription drug3.39g, Zhenqifuzheng particle group dose is0.1g),1times/day. Model control group and blank control group were given equal doses of 0.9%saline. To ensure that the dose does not cause damage to the stomach inmice, oral administration of fasting and water deprivation for2hours, oraland Zhenqifuzheng Aetna group were given particle heating.2.4Outcome measures:Mice in each group were observed daily hair, activity,diet, two stools and death, respectively, in experiment1and day dosingexperiment10days after the end of24hours measured body weight of mice ineach group, body weight was observed in mice before and after treatment changesin the first10days of administration experiment ends24hours later, miceenucleation method using blood samples were collected using a centrifuge tubecontaining heparin blood was collected peripheral blood of mice by ELISA IL-2,IL-4, IFN-γ levels. After the lastadministration24h, angular vein plexus from mouse blood samples were collectedto detect peripheral blood IL-2, IL-4, INF-γ levels.2.6Antai oral liquid on H22tumor immune related cytokines in rats:tumor-bearing mice in each group daily intraperitoneal injection ofcyclophosphamide20mg/kg, experimental groups gavage for10consecutive days.After the last administration24h, angular vein plexus from mouse blood sampleswere collected to detect peripheral blood IL-2, IL-4, INF-γ levels.Result:1by Antai Oral Liquid S180mice immune-related cytokines in rats shows thatafter treatment, oral high-dose group Antai, Antai oral high-dose group,Zhenqifuzheng particles serum tumor bearing mice IL-2, IL-4, IFN-γ levelscompared with the model group significantly different (P <0.05), couldsignificantly increase in tumor-bearing mice immune-related cytokine levels, improve immunity in mice that Dutch immune mice significantly increased levelsof cytokines. Antai oral high dose group of mice serum IL-2, IL-4, IFN-γ washigher than Antai oral high-dose group, Zhenqifuzheng particle groups. ThusAntai oral group can significantly increase tumor-bearing mice immune serumcytokine levels.1.1Through this experiment further validates Antai oral inhibit tumorgrowth, Antai oral high dose group of mice tumor mass less than Antai oralhigh-dose group, Zhenqifuzheng particle groups. The experiments on mice in eachgroup general condition of observation, oral medication Hou Antai Group weightwith the model group was significantly increased compared weight, weight changebefore and after treatment with the model group Antai Liquid there are alsosignificant differences, combined with the small daily rat hair, activity, diet,two stools and mortality observed in the model group died during the test exists,but the poor quality of survival of mice hair, activity, diet, compared withthe other two groups of mice will be reduced.2.by Antai on H22tumor-bearing mice oral immune-related cytokines in ratsshows that after treatment, oral low-dose group Aetna Antai oral high-dose group,Zhenqifuzheng particle group of mice serum IL-2, IL-4, IFN-γ levels comparedwith the model group significantly different (P <0.05), could significantlyincrease in tumor-bearing mice immune-related cytokine levels, improve immunityin mice, so that a small tumor mice in significantly increased levels of cytokines.Antai oral high dose group of mice serum IL-2, IL-4, IFN-γ was higher than Aetnaoral high-dose group, Zhenqifuzheng particle groups. Tumor immune related cytokines by CD4+Tlymphocytes involved in a variety of cellular immunity of the body cells secretecytokines, immune-related cytokines by the increase in cell-mediated immunitycan be introduced by the corresponding strengthened, and through this experimentshows: Aetna oral immune associated with increased levels of cytokines, therebypromoting cellular immune response, which can be launched Antai oral liquid onlow cellular immune function is restored.
