Abstract


Object:Shuanghuanglian Injecting(SHLI) as the subjects drugs, through the spectrum toxicogenomics studies (toxicology and fingerprint associated), to clarify the type of allergens and intensity of toxic effects from toxicological level, and eventual establishing the relevance screening methods of allergens and the effect target, to lay the foundation for the Traditional Chinese Medicine Injection preparation process transformation (anaphylactoid ingredients removing or shielding) study.Method:1Study on the suitability of indicators of anaphylactoid reactions induced by SHLIKM mice were injected with different concentrations SHLI,saline and Compound48/80containing0.4%Evans Blue by tail vein. in30min after administration, the positive rate of auricle blue dye and pinna degree of blue dye score of animals in each group was recorded, and evans Blue exudation from the auricle were determined quantitativly.Wistar rats were by tail vein were injected with injection different concentrations of SHLI, saline, Compound48/80and the OVA. For SHLI, saline, Compound48/80group, in30min after administration, the plasma histamine levels and serum blood trypsin and complement levels were determined; For SHLI, saline and OVA group, they were sensitized three times, at10th days after the last sensitization,blood serum IgE levels were determined.2Study on preparation conditions of SHLI subjects sampleThe extraction solvents of different polarity was obtained by the solubility parameter method, then selected11kinds of solvent to extracte different polarity paragraph ingredients group of SHLI, and their fingerprinting and similarity was evaluated.3Study on the spectrum toxicogenomics of anaphylactoid induced by SHLIThe toxic effects of anaphylactoid induced by was examined and evaluated by the homogeneous design, combined with Mathematical model of the spectrum toxicogenomics, then calculating the relationship between the SHLI component and toxicogenomics indicators, to get the spectrum poison parameters.Result:1The experimental results show that all positive animals injected drug Compound48/80appeared different degree of blue dye in binaural, the reaction rate and ears blue dye rate reached100%; SHLI360,720,1440mg/kg group response rates reached respectively80%,90%,100%, and the ear blue dye rate80%,85%,100%, compared to the control group showed a significant difference (P<0.01). The analysis about auricle exudation of Evans blue quantitative showed, the positive group and SHLI360720,1440mg/kg group had significantly increased (P<0.01), compared the control group.The results showed that the concentration of total IgE in the OVA group with118.4μg/mL, compared with the control group was significantly higher (p<0.05) and SHLI group was significantly higher (p<0.05)also, but there not was positive correlation trend significantly with dose. Compared with control group, the histamine concentration with2.49μg/mL in compound48/80group was significantly higher (p<0.05). And the plasma histamine and tryptase concentration in SHLI group had positive correlation trend significantly with dose. The total complement concentration in Compound48/80group with77.15U/mL, was significantly higher (p<0.05) compared with the control group,and in SHLI group was significantly higher (p<0.05)also, but the complement level not increased insignificantly with the dose increased (p>0.05). 2The results show that the fingerprint of composition groups extracted by different polarity segment solvent. with different types of ingredients and content. Ultimately, the largest differences of component group extraction solvent is selected as SHLI extraction solvent.3Results show that with histamine, tryptase, total complement and C3a as testing indicator, combining mathematical model of spectrum toxic and ShuangHuangLian fingerprint spectrum,we get the function coefficient of component and target.Conclusion:By comparing coefficient,that screening out the anaphylactoid ingredients suspicious X1, X4, X5, X8,X10, X12, X14in fingerprint spectrum of350nm and X12, X24, X32of254nm. The ingredients, which can produce allergy as injections, did not sensitizing as oral,that have yet been explained. It is a possibleand that ingredients may cause allergic reactions with the formation of supramolecular. But it just a guess, the specific verification needs to be done in-depth follow-up study.
