Abstract


Objective:To explore the minimum toxic concentration and the toxic mechanism of aconitine on interstitial cells of Cajal(ICC) from murine colon,and to investigate the mechanism of the compatibility of emodin and aconitine on the ICC.Methods:Use the Uniform design and MTT to find out the minimum toxic concenteation of aconitine on ICC; the MTT method was used to determine how the mixture of emodin and aconitine infect the survivial condition of ICC in different proportions; optical microscopy was uesd to observe the influence on cell morphology by aconition, emodin, and the mixture; turbidimetric analysis, cell enzymology mothod and the ELISA were employed to research the affection on cell biomembrane, the cell homeostasis and energy metabolism by the compatibility of aconitine and emodin on the ICC; then to observe the effect on ICC by the mixture of emodin and aconitine, and to discussion the mechanisms of the compatibility of emodin and aconitine.Results:The least toxic density of aconitine on ICC was0.0025%. The toxic concentration of aconitine lead to the number of cells dcreased, the cellular morphology changed, cellular processes broken, emerging many cytoplasmic vacuoles, the cellular refractivity disappeared and the cytoclasis. when the emodin was used with the aconitine, the changes can be lightened in varying degrees.After0.1%aconitine working on ICC for30min, the activity of ACP and the contents of MDA were increased; the contents of [Na+]、[K+]、[Ca2+] were decreased; the activity of Na+K+-ATPase. Ca2+-ATPase、Ca2+Mg2+-ATPase were decreased. After0.005%aconitine working on ICC for30min, the activity of ACP and the contents of MDA were increased; the contents of [Na+]、[Ca2+] were increased, and [K+] was decreased; the activity of Na+K+-ATPase was increased, and Ca2+-ATPase、Ca2+Mg2+-ATPase were decreased. After the different levels of emodin working on ICC for30min, in varuous degree, the activity of ACP and the contents of MDA were increased; the contents of [K+] was decreased, and [Na+]、[Ca2+] were increased; the activity of Na+K+-ATPase、Ca2+-ATPase、Ca2+Mg2+-ATPase were significantly increased.The compatibility of toxic concentration of emodin and conitine could aggravate the toxicity in ICC of colon. The mixture of aconitine and antional concentration of emodin could lighten the up changes in diffentent levels, then the emodin can antagonism the toxic effect of aconitineConclusions:0.005%and0.1%aconitine had significant toxicity on ICC. They directly damaged the cellular morphology, and also could cause damage of the biomembrane, disturb the internal environment, obstacle to the energy metabolism. When aconitine was used with emodin in different consistency, the changes caused by aconitine could be lightened, then the toxicity reducing and the efficacy enhancing. The mechanism on ICC probably conducted in following way; the emodin can inhance the activity of Ca2+-ATPase and Ca2+Mg2+-ATPase, and improve the uptake ratio of [Ca2+] witch intussuscepted by cell membrane、sarcoplasmic reticulum、endoplasmic reticulum. Then the increase of [Ca2+], witch caused by aconitine damaging the biomembrane, can be inhibtied; the cells were to be kept the minimal level of [Ca2+]; the membrane was to be steady; the ICCs were promoted to produce the slow wave pacemaker current; the normal physiological activities of the ICCs can be restored.
