BackgroundMetabolic syndrome (MetS) is a variety of metabolic components abnormalaccumulation of pathological state,the metabolic syndrome is characterized bycardiometabolic risk factors including insulin resistance, obesity, dyslipidemia, andhypertension. Metabolic syndrome will accelerate the occurrence of coronary heart diseaseand other atherosclerotic cardiovascular disease, development and death. Oxidative stress isclosely related to diabetes, hypertension, artery atherosclerosis (AS), a variety of diseases.Recent studies have found that the hydrogen gas is an important physiological regulator ofthe factor, it is possible to reduce oxidative stress, may become a novel biologically activemolecule for treating a variety of diseases. In the biomedical field, many studies have shownthat by inhalation of hydrogen, a saturated solution of hydrogen injection or drink can beimproved by a variety of disease states or correcting abnormal variety of indicators, such asthe improvement of the lack of type II diabetes, chronic hepatitis, acute oxidative stress, andcentral brain the blood reperfusion injury, and our team recent studies have also found thatthe lipid-lowering effects of hydrogen on the high-fat diet-fed Syrian golden hamster.However, the influence of hydrogen on the blood lipid and lipoprotein levels are still yet tobe determined.ObjectiveThe purpose of this experiment is to study the hydrogen on the metabolic syndromeserum lipoprotein composition and biological activity of hydrogen-containing drinkinghydrogen-rich water (0.91.0L/d).MethodsSelect a community to meet the conditions of metabolic syndrome20volunteerpatients, drinking hydrogen-rich water900ml per person per day, respectively taken fasting blood after drinking water0and10week.the enzymatic and fast protein liquidchromatography (FPLC)analysis, detection of plasma total cholesterol (TC), triglyceride(TG), high density lipoprotein cholesterol (HDL), low density lipoprotein cholesterol(LDL),very low density lipoprotein(VLDL)and glucose (GLU) level; Malondialdehyde(MDA) kit, the kit of superoxide dismutase (SOD) and PON-1activity determination ofplasma antioxidant; determination of plasma and blood lipids anti-inflammatory function byELISA; To using cryogenic speeding centrifugation were separated out of VLDL, LDL andHDL respectively; lipoprotein SDS-PAGE electrophoresis analysis of ApoE in the VLDL,ApoB-100in the LDL,ApoA1in the HDL; By Western blot analysis plasma ApoE,ApoB-100and ApoA1protein content; through the protection of HDL suppressionCu2-induced LDL oxidation degree to detect the antioxidant function of HDL; reversecholesterol transport function of the experimental detection HDL cholesterol lipid-ladenmacrophages in charge overflow; protective effect on endothelial cells by monocyteadhesion assay HDL and LDL on endothelial cell stimulation.ResultsSerum analysis showed that consumption of H2-rich water for10weeks resulted indecreased serum total-cholesterol (TC) and low-density lipoprotein-cholesterol (LDL) levels;HDL, SOD levels have increased; SDS-PAGE analysis of apolipoproteins, ultracentrifugallyisolated from serum, revealed a marked decrease of apolipoprotein B100(ApoB-100) inLDL. Besides, we found H2significantly improved high-density lipoprotein (HDL)functionality assessed in four independent ways, namely (i) protection against LDLoxidation,(ii) inhibition of TNF-αinduced monocyte adhension to endothelial cells,(iii)stimulation of cholesterol efflux from macrophage foam cells,(iv) protection of endothelialcells from TNF-α induced apoptosis. Further, we found consumption of H2-rich waterresulted in an increase in antioxidant enzyme superoxide dismutase and a decrease inthiobarbituric acid-reactive substances in whole serum and LDL.In conclusion,supplementation with H2-rich water appear to decrease serum LDL and ApoB-100levels,improve dyslipidemia injured HDL functions, and reduce the oxidative stress and may havea beneficial role in prevention of potential metabolic syndrome.Therefore, these resultsshow that hydrogen can be reduced oxidative damage, may be provided on the metabolicsyndrome, metabolic syndrome generated by the oxidative stress in patients inhibition andtherapeutic effects.Conclusions1. H2-rich water significantly improve the metabolic syndrome (MetS) lipid levels, decreased plasma TC and LDL levels;2. H2-rich water significantly reduce the major protein component of the MetS patientswith LDL level of ApoB-100;3. H2-rich water significantly improved in patients with MetS plasma HDL functions,including anti-oxidation, anti-adhesion, anti-apoptotic, and the promotion of reversecholesterol transport function;4. H2-rich water improved the MetS patients the degree of oxidation of the LDL andthe stimulation of endothelial cells.
