Abstract


Objective:A case match control study were carried out to aim at exploring the relationship between the polymorphisms of Vitamin D Receptor (VDR) gene and the susceptibility to pulmonary tuberculosis (PTB) among the populations in Ningxia Hui Autonomous Region (NHAR), P.R. of China.Methods:All the confirmed PTB cases, who had been diagnosed according to WHO TB-diagnosis criteria, without any other co-infectious condition, in different local TB clinics in NHAR, were recruited under free-will in2010, and also the healthy match controls, who were selected by the criteria of the same sex, nationality, similar age and residential area to those cases but without any health condition. All the participants were interviewed by TB clinic staff with a questionnaire sheet regarding to the demographic information and the factors related to the diagnosis, treatment and TB transmissions. The participants were required2-5ml venous blood in EDTA tube, then stored at-20℃within30minutes post sampling before DNA extraction. After genomic DNA extraction, the detection of two SNPs (rs731236、 rs2228570) in VDR gene were carried out by use of the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The data were entered into the SPSS16.0software package. After the effective value of population data was tested by use of Hardy-Weinberg law method, a multiple regression analysis were used for the different values between SNPs in different gene presentation models. The significant value was determined in95%confidential intervals with P<0.05. ResuLts The distributions of the two SNPs of VDR gene among the993PTB cases and880health match controls were determined in effective value (P>0.05) that can represent features in local populations. Therefore, the outcomes of the comparisons of PCR-RFLP on two SNP data between cases and controls can be used for further analysis. The RFLP data analysis by use of the multiple logistic regression model has been found the significant differences in the gene models of VDR (TT+Tt), VDR (FF-ff) between cases and controls generally, which represented TB infection susceptibility increasing (VDR-TT) in males and decreasing (VDR-FF+ff) in Hui Islamic populations.Conclusion:VDR gene polymorphisms have been fond to be association with either the susceptibility (VDR-TT+Tt), particularly in males, or the resistance (VDR-FF+ff), particularly in Hui Islamic populations to the human PTB infection among Ningxia local populations.
