Objective: To explore whether the surgical delay of the subdermal vascular networkskin graft and subdermal injection of VEGF can improve the survival rate of skingraft;2. Observed the diameter and cross-sectional changes after the operation ofthe subdermal vascular network;3.Discussing the number changes in the averagemicrovascular using the growth factor (VEGF) and the roll played by theadrenomedullin (ADM) in the mechanism of formation of subcutaneous bloodvessels in the skin graft delay.Method: Rats with subdermal vascular network skin graft delay model,intradermal injection of VEGF model, and the superposition model were created toobserve the changes of survival rate,blood vessel diameter and the cross sectional areaafter the skin graft; The delay model skin graft within the organization VEGF andADM qualitative and quantitative analysis were carried out using the enzyme-linkedimmunosorbent assay (ELISA)in the organization of skin delay model to detectwhether the changes of the expression of ADM and the VEGF in the skin graft surgerywere correlated with the changes of subdermal vascular network using the statisticalanalysis.Result:1.The survival rate of growth factor group,the the delayed group and theoverlay group was significantly higher than that of the saline group (P <0.05);there is no significant difference between the delayed group and the growth factorgroup(P>0.05). The superimposed group were significantly different than that ofother experimental group (P <0.05);2、Comparing with the base value,the levels ofVEGF in the delay model after3days skin graft increased significantly but sigifcantlyhigher than that of7/14d,(P <0.05)and no significant differences of14days with thebase values.3、The expression of ADMmRNA were analysised using the RT-PCR inthe wound tissue and detected using the double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) in the peripheral blood and the wound tissue.Theresults shows that there was no significant difference with the chip average vesseldiameter(MVD) of corresponding delay time interval.Conclusion:1、Intradermal inection of VEGF on Rat can promote local vascularendothelial growth, with the biological effects of promoting the formation of newgranulation;2.Both the Model of subcutaneous injection of vascular endothelial growthfactor (VEGF) and skin graft delay can effectively improve the survival rate of the skingrafts of subdermal vascular network, but the mechanism of the two wasdifferentgrowth factor (VEGF) can further improve the survival rate of skingraft;3.Compared with the saline control group, the skin graft with subdermal vascularnetwork has a higher survival rate;It came to the conclusion that the VEGF played animportant role in t he delaying process of skin graft and the process ofvascularization;3.The results suggest that although there are changes of ADMmRNAexpression in the flap tissue after the graft trauma in two weeks, the content of theperipheral blood and Local tissue flap were detected by enzyme-linked immunoassay(ELISA) shows no significant changes of the ADM concentration and to a certainextent that there is no significant correlation with vascularization of subdermal vascularnetwork.
