Expression of Notch1/Delta-like4on Peripheral Lymphocytes of Renal Transplantation Recepients and the Relationship with Th17

Abstract


Objective To study the changes in expression on peripheral lymphocytes and investigate the role of Th17cells in acute allograft rejection after renal transplantation.Methods20Patients suffering early aeute rejection after receiveing a renal transplantation(<1month) in2012.1-2013.12are arranged as acute rejeetion(AR) group.12patients with post-operation acute tubular neerosisare arranged as ATN group.20patients with normal post-operation renal function are arranged as control group. Flow cytometry used to detect the expression of Notchl/Delta-like4and the percentage of Th17cell on peripheral lymphocytes of the patients before operation, and days1,3,7,14after transplantation, in addition, these target were also detected when AR and ATN occurrene and days1,3,7,14after theropy. Notch1/Delta-Iike4and the percentage of Th17cell before operation, after operation and after morbidity are compared.Results Preoperative Notchl/Delta-like4and the percentage of Th17cell among AR group, ATN group and control group did nnot show difference (P=0.546; P=0.714; P=0.153). Post-operativel Id,3d Notchl/Delta-like4and the percentage of Th17cell were not difference among3group either (P=0.161; P=0.169; P=0.306). However they showed significant difference on post-operative7d,14d (all p=0.000). Notchl/Delta-like4and the percentage of Th17cell of AR group was higher than that of control group and ATN group on post-operativel7d,14d (all p<0.05), but they were not different between control group and ATN group both on Preoperative and Post-operativel (all p>0.05). Notchl/Delta-like4and the percentage of Th17cell on post-operative1d was higher than preoperative ones on every group(P=0.000). They level on post-operative3d deereased, after7d ATN and control group reached preoperative level(all p>0.05), however, they level increased in AR group on post-operative7d,14d (P=0.000vs preoperativeleve). Notchl/Delta-like4and the percentage of Th17cell of AR group was signifieantly higher than that of ATN group on1d after morbidity and on post-treatmentld,3d,7d (all P=0.000). Notehl/Delta-like4and the percentage of Th17cell of AR group were higher than preoperative ones (P=0.000). After anti-rejeetion, they level decreased continually and reached preoperative level on14d (P=0.172;P=0.139;P=0.841). The ATN group was also higher than preoperative ones on1d after morbidity (P=0.000). After therapy. Notehl/Delta-like4and the percentage of Th17cell decreased on3d (all p>0.05).Notehl and Delta-like4expression on peripheral blood lymphocytes showed significant positive correlation (r=0.968, P=0.000), Notehl expression and percentage of Th17cell, Delta-like4expression and percentage of Th17cell also showed significant positive correlation (r=0.893,P=0.000; r=0.872,P=0.000).Conclusions When acute rejection occurred after renal transplantation, Notchl/Delta-like4expression on lymphocytes increased significantly. Examination of Notehl/Delta-like4expression on lymphocytes is helpful to diagnosis of acute rejection. The D114-Notchl signaling and Th17cell play an important role in acute rejection, selective targeted therapy against Notch may be an important tool to treat acute rejection.
