LIF Promotes the Expression of GP130mRNA on Endogenous Neural Stem Cells in the Mouse Model of PD

Abstract


[Purpose]To detect the re-expression of endogenous neural stem cells (ENSCs) in the areas of substantia nigra, striatum, hypothalamus on the brain of6-hydroxydopamine (6-OHDA) PD mouse model, and declose the expression of gp130mRNA of ENSCs following Leukemia Inhibitory Factor (LIF) administer ation.[Methods]96mice at8-week age were divided into12groups,8mice in each group. To develop a mouse model, mice in all groups accepted three-dimensional brain stereotactic injection of6-OHDA excepted the control group, these groups were ended at the timepoints of1week,2week and3week.1. Mice in all groups accepted neuro-function tests which include Bar Grab test, Rotarod Test, and Tremor analysis, the data was statistically analyzed.2. The re-expression and proliferation of ENSCs in the mouse brain were studied using immunohistochemistry method before and after LIF administration.3. RNA from substantia nigra, striatum, hypothalamus of the hemi-left and hemi-right mouse brain was extracted, the GP130mRNA relative expression was measured using semi-quantitative RT-PCR method.[Results]1. The variance analysis of Bar Grab test, Rotarod Test and Tremor analysis shows that there is no statiscally difference in the1-week groups of Sham, LIF and PD compared with those groups at2-week timepoint (P>0.05). A statistical difference (P<0.05) was found in the LIF group compared with Sham and PD groups at3-week timepoint, and there was no differ ence(P>0.05) in Bar Grab test at LIF and PD groups compared with contr ol (CON) and Sham groups.2. The re-expressed ENSCs distributed around the injured sites were detected in the mouse brain at3-week groups of LIF, PD groups excepted the CON and Sham groups.3. Semi-quantitative RT-PCR:There was no significant difference (P>0.05) in the expression of GP130mRNA at the timepoints of1-week and2-week in all groups. At the timepoint of3-week, a statistical difference was detected in LIF group compared with that of Sham and PD groups (P<0.05), and same pattern was found in the CON group (P<0.05).[Conclusion]LIF is a neurotrophic factor which is able to promote the re-expression of ENSCs, the function recovery of the mice administrated by LIF is significantly increased compared with those in PD and Sham groups. LIF may paly a key role in promoting the expression of GP130mRNA and increasing the re-expression of ENSCs and is a promising way to effectively re-build the degenerated neural cells.
