Objective By immunohistochemistry, we tried to investigate the relationship between the expression level of Ki-67and folate receptor a (FR a) and some clinical factors of pituitary adenomas (age, gender, size, endocrinal types, invasiveness, etc). We also tried to find and confirm some factors to predict the invasiveness of pituitary adenomas. We explored a factor that is specific to certain endocrinal types of pituitary adenomas, in order to make a solid theoretical basis for further targeted diagnosis and treatment of pituitary adenomas.Method We collected51pituitary adenoma samples during operations from the patients of neurosurgical department of PUMC, from November2008to November2009. All the samples were stored in liquid nitrogen. All the cases had detailed clinical and pathological information. According to certain criteria, the cases were divided into invasive group and non-invasive group. By immunohistochemistry of frozen tissue sections, we analyzed the expression level of Ki-67and FR a of each sample. We then used statistical methods to compare the expression level of the two biomarkers and some relevant clinical factors, like invasiveness, age, gender, endocrinal types, course, and mean diameter of tumors.Result The expression levels of Ki-67were significantly different between invasive group and non-invasive group (P=0.007<0.05). The expression of Ki-67had no significant relevance with age, gender, endocrinal type, course, repeated operation or not (P>0.05), but it was strongly related with the mean diameter of tumors (P=0.00<0.01, correlation coefficient=0.663). FR a expression levels were significantly different among different endocrinal types of pituitary adenomas (P=0.001<0.05). Positive staining appeared only among NF pituitary adenomas, and the positive rate was55.2%. For NF pituitary adenomas, FR a expression was much higher in invasive group, than in non-invasive group (P=0.026<0.05). The expression of FR a had no significant relevance with age, gender, endocrinal type, course, repeated operation or not, mean diameter of tumors and the expression of Ki-67(P>0.05).Conclusion Ki-67can be a biomarker to predict the invasiveness of pituitary adenomas, and was relevant with tumor size. FR a was positively expressed only among NF pituitary adenomas, and was related with invasiveness of NFPA. This research directed further targeted diagnosis and treatment of pituitary adenomas.
