Abstract


Objective:The purpose of this test is to analyze the expressions of pituitary tumor transforming gene in normal pituitary, invasive pituitary adenomas and noninvasive pituitary adenomas, respectively, through the immunohistochemical method, to explore its relationship with the occurrence, development and invasiveness of pituitary adenomas, and to lay the theoretical basis for the diagnosis of pituitary adenomas, the judgment of clinical prognosis and the selection of therapeutic schedule.Materials and methods:Randomly select40cases of pituitary adenoma specimens through surgical excision (transsphenoidal surgery or craniotomy) by the Neurosurgery of Central South University Xiangya No.2Hospital from January2012to December2012, and they were confirmed as pituitary adenoma tissues by pathological examinations, with at least100mg of pathological tissues available for the studies. With reference to Handy-Wilson and Knosp classification, there are22cases of invasive pituitary adenomas and18cases of non-invasive pituitary adenomas; including4cases of micro adenomas,29cases of large adenomas and7cases of huge adenomas. There are11cases of functional adenomas and29cases of non-functional adenomas. With7cases of disease-free pituitary tissues as control group, the specimens are derived from the autopsy, and they are also confirmed as disease-free pituitary tissues by pathology. Immunohistochemical SP method is used to detect the expressions of PTTG in normal pituitary tissues, noninvasive pituitary adenoma tissues and invasive pituitary adenoma tissues. The multi-function true color pathology analysis system is adopted for immunohistochemical detection, with the nucleus and (or) cytoplasm developing brownish-yellow granules as the criterion, to select the area with more positive cells for casual inspection of five high power fields (×400) and analysis of the percentage of positive cells accounting for the total number of cells. Analyze the expressions of PTTG in normal pituitary, invasive pituitary adenoma and noninvasive pituitary adenoma tissues.Results:1. The relationship of PTTG expression with the occurrence of tumor:7cases of normal pituitary tissue specimens and40cases of clinical pathology specimen with pituitary adenoma tissues are collected, and the immunohistochemical method is used to detect the expression of PTTG in each specimen. In40cases of specimens with pituitary adenoma tissues, there are a total of33cases of positive staining specimens, accounting for82.5%; In7cases of specimens with normal pituitary tissues, there is only1case developing weak positive staining, and all the rest are not staining. By the chi-square test, the PTTG expression level between the pituitary adenoma tissue and normal pituitary tissue has highly significant difference (P<0.01).2. The relationship of PTTG expression with invasive pituitary adenoma:In18cases of noninvasive pituitary adenoma tissues, there are6cases of specimens without staining,3cases of specimens with weak positive staining,8cases of specimens with moderately positive staining and1case of specimen with strong positive staining; Whereas, in the invasive pituitary adenoma tissues,1case of specimen without staining1case of specimen with weak positive staining,3cases of specimens with moderately positive staining and17cases of specimens with strong positive staining. By the chi-square test, the PTTG expression level between noninvasive pituitary adenoma tissue and invasive pituitary adenoma tissue has significant difference (P<0.05).Conclusions:1. PTTG expression rate difference between pituitary adenoma tissue and normal pituitary tissue is highly significant, indicating that the occurrence of pituitary adenoma is closely associated with the roliferation of PTTG. 2. PTTG expression rate difference between the invasive pituitary adenoma tissue and noninvasive pituitary adenoma tissue is significant, indicating that its expression is associated with the invasive degree of pituitary adenomas.3. The overexpression of PTTG may be an important mechanism for the occurrence and development of pituitary adenoma, so it may be taken as an indicator for the evaluation of prognosis of pituitary adenomas and may become a new target for the treatment of pituitary adenomas.
