Objectives:To explore the effect of inhibiting glial activation on the rat retinal ganglion cells (RGCs) loss in acute intraocular hypertension.Methods:1、36adult SD rats were divided randomly into control group and experimental group, experimental group was divided into6h group; Id group;3d group;5d group and7d group according to the time after treatment. Acute ocular hypertension model was established by increasing intraocular pressure (IOP) through anterior chamber infusion. Choose one eye for treatment, while the other eye remained sham-treated. Change of retina was observed before or after the surgery. The levels of Thymosin-1(Thy-1) in every group were detected by immunohi-stochemical (IHC).2、18SD rats were divided randomly into blank control group experimental group and control group. The acute ocular hypertension model was established by increasing IOP through anterior chamber infusion. After the establishment of model, SD rats were treated with AG1478(epidermal growth factor receptor inhibitor) in the drinking water (6mg/kg/d), control group was treated with physiological saline of the same volume. Animals were allowed to survive for7d. Histological changes in retina and the expression of RGCs were observed by IHC.Results:1、Each layer of the retina became not clear, Thy-1expression and positive cells of ganglion cell layers gradually reduced after acute IOP increased.2、The mean optical density of Thy-1positive cells in blank control group, model group and control group were:143.6667±4.0415;139.0322±1.7340and101.1023±2.0001. No statistical difference between model group and blank control group (P>0.05). The expression of Thy-1positive cells in blank group was much higher than that in controls (P<0.05).Conclusions:1、Through anterior chamber infusion can obtain pathology change that is similar to acute angle-closure glaucoma.2、Inhibiting glial activation has an protective effect of RGCs on rat with acute intraocular hypertension.
