Abstract


Objective To study the effect of curcumin on cultured choriocarcinoma JEG-3 cell growth and cell survivin transcription of mechanism of action of curcumin in JEG-3 cell proliferation and apoptosis in the clinical treatment of choriocarcinoma application prospects. JEG-3 cells cultured in vitro for the study, with different concentrations (2.5μmol / L, 5 micromol / L, 80μmol / L, 160μmol / L) of curcumin treatment as the experimental group, curcumin-free culture medium The treated cells as a negative control group. 24, 48 and 72 hours after detection tetrazolium blue method (MTT) cell activity, Annexin V FITC / PI double staining flow cytometry and RT-PCR were detected 20 micromol / L, 40 micromol / L and 80μmol / L curcumin for 48 hours after the incidence of apoptosis and survivin transcriptional level. Curcumin JEG-3 cell growth inhibition and concentration-and time-dependent: the same duration of action, curcumin treatment groups and the control group were statistically significant differences (p lt; 0.05), and the inhibition increased significantly with the increase of the concentration of curcumin (r = 0.913, P lt; 0.05); the curcumin certain concentration (concentration ≥ 10 micromol / L) on the JEG-3 cell line with the extension of the duration of action to strengthen significantly (r = 0.751, p lt; 0.05); curcumin IC50 of 24, 48 and 72 hours in JEG-3 cells and 87.96μmol / L 37.30μmol / L and 24.99μmol / L,. Curcumin induced apoptosis in JEG-3 cells, early apoptosis rate increased with increasing concentration (r = 0.966, p lt; 0.05): 0 mol / L, respectively, 20 micromol / L, 40 micromol / L, 80μmol / L curcumin on JEG-3 cells and 48 hours after the early apoptosis rate as follows: (0.83 ± 0.79)%, (10.54 ± 2.84)%, (22.38 ± 6.99)%, (42.65 ± 6.71)%, among the three groups compared to the differences were statistically significant (p lt; 0.05). Curcumin reduced survivin expression decreased with increasing concentration (r = -0.900, p lt; 0.05): 0 mol / L, respectively, 20 micromol / L, 40 micromol / L, 80μmol / L curcumin JEG -3 cells after 48h, survivin mRNA relative expression levels were: 0.8032 ± 0.0380,0.4057 ± 0.0206,0.3129 ± 0.0371,0.1623 ± 0.0257, compared to the differences were statistically significant (p lt; 0.05) among the groups. Conclusion Curcumin JEG-3 cells, inhibition of proliferation and induction of apoptosis, which may be related to reduced expression of survivin.
